| Objective:Our aim is to investigate the relationship between ERαand ERβmRNA expression levels in different brain regions(marginal,hypothalamus,limbic cortex and hippocampus)and the pathogenesis of PMS with liver-qi depression,as well as to study the effects of Jingqianshu granule.Methods:We made the model rats of PMS with liver-qi depression with the methods of resident intruder paradigm and emotional stimulation multiple factors respectively, confirmed the estrum of rats by observing the vagina cell shape,and performing behavioral evaluation at the acceptance period and the non-acceptance period.We detected the expression levels of ERαand ERβmRNA in different brain regions of rats in groups including the normal group,the model 1 group(resident intruder paradigm model group), the model 2 group(bound model group)and treatment group by using a semi-quantitative RT-PCR gene amplification technology.Results:We found out the two modeling methods could simulate women's symptoms of PMS with liver-qi depression effectively.There was no significant difference(P>0.05) between the two methods in open-field experiment.The expression levels of ERαand ERβmRNA in hypothalamus,hippocampus,marginal and cerebral cortex of model rats were higher compared with the control group(P<0.05);while in the Jingqianshu graulne group,the expression levels of ERαand ERβmRNA were lower than model group(P<0.05), and have no difference with the control groupConclusion:Resident intruder paradigm as a psychosocial stress model method can better simulate the process of clinical emotional abnormalities that triggered PMS with liver-qi depression,but it's disadvantage is low success rate(the success rate is only 22.5%),and the stability of the model also need to be further examined.The expressions changes of the ERαand ERβmRNA are in close relation to PMS with liver-qi depression.At the same time, the Jingqianshu granule can down-regulate the ERαand ERβmRNA expression levels of model rats in different brain regions,which maybe one of the mechanism to treat PMS with liver-qi depression. |
