Objective:To investigate the GHR exon 3 polymorphsim and its relationship with the metabolic profile in Chinese obesity children.Subject and Methods:409 obesity/overweight children and 206 normal weight children were recruited.Body weights and heights were measured,body mass indexes were calculated,obesity degree were evaluated using IOTF standard.Genomic DNA was extracted from their peripheral blood leukocytes,GHR exon3 gene polymorphism were determined using PCR method.Fasting glucose,insulin and lipid profile were analysed, and HOMA-IR and ISI were calculated using homostais model.All the data were analysed by SPSS 12.0 software.Result:1.GHR exon3 gene polymorphism distribution in obesity and normal control group:The frequency in obesity and overweight group of the three gene types d3/d3,f1/d3,f1/f1 is 7.3%,7.9%and 4.8%;The frequence in control group of the three gene types d3/d3,f1/d3, f1/f1 is 4.8%,17.5%,77.7%.The frequence of d3 gene in obesity group is significantly higher than that of the control group(χ~2=10.66,p=0.005).2.GHR exon3 gene polymorphism and its relationship with metabolic profile in obesity group:The bioactivity of GHRd3 isoform is about 30%higher than GHRf1,so the d3/d3 and f1/d3 genotypes were combind as d3 group.In the obesity group,144 subjects were d3 genotype,and 265 were f1/f1.There existed no statistical significant difference in age (t=-0.21,p=0.83),heigh(t=-0.20,p=0.84),weight(t=-1.73,p=0.17),and sex ratio between the two genotypes(χ~2=0.11,p=0.74).The BMI(t=-2.4,p=0.02),fasting insulin(t=-2.91, p=0.004),HOMA index(t=-5.11,p=0.00),total cholesterol(t=-2.34,p=0.02), triglyceride(t=-2.16,p=0.032) of the d3 group was significantly lower than of the f1/f1 group.The insulin senstive of the d3 group is significantly higher than that of the f1/f1 group(t=4.37,p=0.00).3.GHR exon3 gene polymorphism and its relationship with metabolic in normal control group:In the control group,46 subjects were d3 gene,and 160 were f1/f1.There existed no statistical difference in BMI(t=1.03,p=0.30),fasting insulin(t=0.25,p=0.80),HOMA index(t=-0.08,p=0.94),insulin sensitive index(t=0.59,p=0.56),total cholesterol (t=-0.23,p=0.82) and triglyceride(t=-0.20,p=0.84) between the two genotypes.Conclusion:The GHR d3 polymorphism has no significant effect on children's obesity development,but GHR d3 genotype can preventing obesity children from developing into metabolic syndrome via increasing insulin sensitivity. Objective:To identify the association of the IGF1 Gene SNP rs5742694G/T with adult height and body mass index(BMI) in young Chinese adults.Patients and Methods:1000 young adults including 500 male and 500 female from a college were recruited.Body heights and weights were measured,and BMI were calculated.Genomic DNA was extracted from their peripheral blood leukocytes.SNP rs5742694G/T was determined by SnaPshot.The genotype and phenotype correclation were analyzed by SPSS15.0.Results:1.SNP rs5742695 existed three genetypes,TT,TG and GG.The most frequent genetype is TT,and GG is rare both in male and female subtypes.The mean height of each genotype is,for male,TT 172.10cm,TG 172.74cm,GG 172.12cm,there existed no significant statistical difference among the three genotypes(one-way ANOVA analysis p=0.57).The mean height was TT 160.19cm,TG 159.21cm,GG 159.74cm in female group,there existed no significant statistical difference in the three genotypes(one-way ANOVA analysis p=0.73).2.The extreme discordant phenotype design was used to find the height difference between the three genotypes.100 shortest male(H≤170cm) and 111 tallest female (≥177cm) were selected,comparing their gene constituent ratio,and find there existed no statistical significant between the gene constituent composition between the shortest and tallest groups(p=0.70).92 shortest female(H≤155cm) and 108 tallest female(≥165cm) were selected,comparing their gene constituent ratio,and find there existed no statistical significant between the shortest and tallest groups(p=0.47).3.There existed no statistical significant BMI difference among the 3 genotypes (p≥0.05).There also existed no statistical significant in the geneconstituent ratio between the thinnest and fattest males and females(p≥0.05).4.There revealed no significant association among the 3 SNP genotypes and heights or BMI in both sexes using superposition linear regression model analysis.Conclusion:The IGF-1 intron 3 SNP rs5742695 G/T polymorphism showed no significant effect on the adult height and body mass index in current Chinese young people. Objective:To identify the association of the ESRαGene SNP rs2179922G/A with adult height and body mass index(BMI) in young Chinese adults.Patients and Methods:1000 young adults including 500 male and 500 female from a college were recruited.Body heights and weights were measured,and BMI were calculated.Genomic DNA was extracted from their peripheral blood leukocytes.SNP rs2179922G/A was determined by SnaPshot.The genotype and phenotype correclation were analyzed by SPSS15.0.Results:1.SNP rs2179922 exists three genotypes,TT,TG and GG.The most frequent genotypes are GG and GA,and AA is rare both in male and female subtypes.The mean height of each genotype were,for male,TT 172.22 cm,GA 172.23cm,AA 172.03 cm,there existed no significant statistical difference in the three genotypes(one-way ANOVA analysis p=0.88).The mean height was TT 160.34cm,TG 159.74cm,GG 160.30cm in female group,there exixted no significant statistical difference in height of the three genotypes(one-way ANOVA analysis p=0.43).2.The extreme discordant phenotype design was used to find the height difference between the three genotypes.100 shortest male(H≤170cm) and 111 tallest female (≥177cm) were selected,there existed no statistical significant difference in the gene constituent ratio between the shortest and tallest group(p=0.43).92 shortest female(H≤155cm) and 108 tallest female(≥165cm) were selected,there existed no statistical significant difference in the gene constituent ratio between the shortest and tallest group(p=0.37).3.There existed no statistical significant BMI difference among the 3 genotypes(male p=0.59,female p=0.29).There also existed no statistical significant difference in the gene constituent ratio between the thinest and fattest group by extreme discordant phenotype design study(p≥0.05).4.There revealed no significant association among the 3 SNP genotypes and heights or BMI in both sexes using superposition linear regression model analysis.Conclusion:The ESRαGene SNP rs2179922G/A polymorphism showed no significant infulence on adult height and body mass index in young Chinese people.
