| Objective To explore the therapeutic alliance of Adeno-associated virus-based angiostatin(AAV-AS) cooperated with Cytoxan(CTX) on tumor growth, angiogenesis and tumor apoptosis of subcutaneous gliomas in rats.Methods Subcutaneous C6 glioma animal modes were established.The rats were randomly assigned into 4 treatment groups after tumors reached about 250~280mm~3 after 7 days.A:saline control group;B:CTX chemotherapy group;C:AAV-AS group;D: therapeutic alliance group.Perpendicular diameters of tumors were determined by measured two every 3 days and volumes were calculated.The rats were killed after 22 days.The expression of AS protein was performed to detect by immunohistochemistry in situ.Tumor vascularity was performed to assess by CD31 staining.Apoptosis of the tumor cells was performed by TUNEL analysis.Result The AS transcription were detected from AAV-AS and therapeutic alliance groups,which can confirm the achievement of genic transduction.The volumes of tumors in group D(1098.320±70.880mm~3) were significantly reduced(P<0.05),when compared with groups A(2060.339±82.028 mm~3),B(1763.867±124.237mm~3) and C (1229.910±141.646mm~3),therapeutic alliance can inhibit the volumes of gliomas. The vascularity of tumors in group D(8.364±3.572/0.24mm~2) were significantly reduced(P<0.05),when compared with groups A(19.810±3.423/0.24mm~2),B (17.737±4.658/0.24mm~2)and C(12.631±2.359/0.24mm~2),the therapeutic alliance can inhibit the vascularity of gliomas.The apoptotic index of group D(3.886±0.381) were significantly increased(P<0.05),when compared with group A(1.442±0.776) and B(1.719±0.344),the therapeutic alliance can induce the apoptotic of glioma cells indirectly.Conclusion The antiangiogenesis gene therapy cooperated with chemotherapy can inhibit tumor growth by inhibiting tumor vessel generations.This Therapeutic alliance has synergic effect,and can retrieve the shortage of each one. |
