| Objective In order to study lymphangiogenesis of colorectal carcinoma,the expression of vascular endothelial growth factor C(VEGF-C) and signal transducers and activators of transcription 3(STAT3) were assayed,their relationship and relationship with clinopathologic features were analysed,as well as their effects to invasion and metastasis of CRC were demonstrated.Meanwhile,the regulating of controlling genes of lymphangiogenesis were determined using gene microarray as a basis of deep research of lymphangiogenesis machanism.Methods The levels of VEGF-C,STAT3 mRNA transcription and protein espression of adenocarcinoma and surgical margin tissues gotten from 60 cases with CRC were determined by real-time PCR and immunohistochemical strain,separately,the samples coming from 2 cases whose tumor tissues were with high lymphatic vessel density and 2 cases with low LVD were examinated by functional microgene array.Results The levels of VEGF-C and STAT3 of tumor tissues were higher than that of surigical margin in either mRNA transcription or protein expression.A significant positive relation between STAT3 expression and VEFR-C expression was showed.There were a statistics difference in the differentiation of tumor,the depth of tumor,lymphatic node metastasis as well as Dukes stage of colorectal cancer in both mRNA and protein of VEGF-C and STAB.The different expresses of VEGF-C and STAT3 in different age,gender,primary tumor location and pathologic type were not detected.Comparing results of gene microarrgy of high LVD tumor and low LVD tumor up/down regulation more than 5 folds in those genes,it is well understanding of relationship between VEGF-A and VEGF-C and hypothesis that VEGFA is one of up grade regulation to VEGF-C was probably established.Conclusions The higher expression of VEGF-C in colorectal cancer than that in surgical marginal tissue demonstrate that VEGF-C is very important molecular to rich formation of lymphatic vessels in colorectal cancer.The positive correlation between STAT3 and VEGF-C in colorectal cancer suggests that STAT3 is important positive regulator of lymphangiogenesis in up grade.The different expression of VEGF-C and STAT3 in different differentiation of tumor,depth of tumor invasion,lymphatic node metastasis and Dukes stages in colorectal cancer indicates that these moleculars could be used in jugement of metastasis and prognosis of colorectal cancer.The lymphangiogenesis in colorectal cancer is the result of multi-genes and multi-channels,and VEGFA may play a key role in lymphangiogenesis of colorectal cancer. |
