Nowdays,obesity has been regarded as a kind of epidemic diseases by WHO,which has a serious influence on the health of human beings,and affects the quality of human life,and causes a rapid increase in medical expenditures.It is also a risk factor for most diseases and can cause a variety of diseases and serious complications.Generally,body metabolic disorder results in occurrence of obesity,which the body energy intake is more than consumption,and remains the stored fat into the body.In recent years,many studies show that obesity is associated with genetic gene,environment,dietary structure and so on. It is one of the global prevalent and independent chronic diseases characterized with fat excess of body.Recently,more and more studies focus on reducing blood lipid of drugs by antioxidation,and a lot of data showed that obesity induced by metabolic disorder has a close relation with redox system.Therefore,to explore the mechanism and prevention measures on obesity has become a main task in continuable development 21st century. Moreover,many clinical and experimental researches confirmed that Chinese herbs had a better anti-obesity effect.It is reported that Gastrodin could scavenge reactive oxygen species(ROS) and protect cell membrane.However,the effects of Gastrodin on free fatty acid(FFA) release and antioxidant activities from isolated adipose tissue in rats and its mechanism are not yet reported.In our experiment,we used gastrodin,an effective monomer of gastrodia rhizome,to observe the effects of gastrodin on biochemical indicators from isolated adipose tissue in rats.As a result,a theoretical foundation is obtained for research of tranditional Chinese herbs with obvious effects but fewer side effects.The experiment was carried out in the Institute of Physiology and Psychology,School of Basic Medical Science,Lanzhou University and Laboratory of Pre-clinical Study for New Drugs of Gansu Province.The healthy SD rats of 250±20g were selected.Krebs solution 2ml with 4%calf serum albumin was added into each group at 37℃with mixed gas of 95%O2 and 5%CO2.After rats were killed by dislocation,the retroperitoneal adipose tissue of 500±20mg was quickly removed and cut into 8~10 pieces in each group. The experiment was divided into eight groups:control group,Adrenaline hydrochloride group(1.0x10-6 mol/L),low,medium and high Gastrodin groups(the concentrations of Gastrodin were 1.0x10-5,1.0x10-6,1.0x10-7 mol/L,respectively),medium Gastrodin plus Verapamil group,medium Gastrodin plus Propranolol group,and medium Gastrodin plus Adenylyl cyclase inhibitor(SQ22,536) group(the final concentration of above inhibitors for the 1.0x10-5 mol/L).Different drugs were added to isolated rat adipose.After incubating 90 minutes,the levels of FFA,malondialdehyde(MDA) and total antioxidative ability(T-AOC) were measured by colorimetric method.ELISA was used to measure the levels of Leptin,TNF-a,IL-6.SPSS 11.5 software was used for data analysis. The experiment rusults were expressed as Mean±SD.The one-way analysis of variance was adopted for comparison among different groups.The main results in the experiment were as follows:1.Compared with control group,Gastrodin dose-dependently increased FFA release, but its effect was weaker than that of Adrenaline hydrochloride.However,compared with medium Gastrodin,the levels of FFA were reduced in medium Gastrodin plus Verapamil, medium Gastrodin plus Propranolol and medium Gastrodin plus SQ22,536 group (P<0.05).2.Compared with control group,T-AOC was raised in Adrenaline hydrochloride group and medium Gastrodin group(P<0.01);compared with medium Gastrodin,T-AOC was reduced in medium Gastrodin plus Verapamil,medium Gastrodin plus Propranolol and medium Gastrodin plus SQ22,536 group(P<0.05).3.Compared with control group,MAD level was reduced in Adrenaline hydrochloride group and medium Gastrodin group(P<0.01);compared with medium Gastrodin,MAD level was raised in medium Gastrodin plus Verapamil,medium Gastrodin plus Propranolol and medium Gastrodin plus SQ22,536 group(P<0.05).4.Compared with control group,the levels of Leptin,TNF-a,and IL-6 did not change(P>0.05).In conclusion,our results indicate that Gastrodin similar to Adrenaline hydrochloride can dose-dependently increase FFA release from isolated adipose tissue in rats.Its mechanism may partly be mediated byβ-adrenoceptor,Varapamil-sensitive L-Ca2+ channel and Adenylyl Cyclase.Gastrodin has antioxidant capacity in the isolated adipose tissue and reduces the extent of lipid peroxidation.Meanwhile,Gastrodin did not affect the levels of Leptin,TNF-a,and IL-6.
