In this paper, pomegranate peel and pomegranate seed were used as raw materials, carry out experiment of purification towards alcohol extract components from pomegranate rind, the structure of the initial identification and determination of its antioxidant and antibacterial activity. Pomegranate seed oil on the physical and chemical indicators for the determination of antioxidant activity, and gas chromatography - mass spectrometry of the fatty acid composition of pomegranate seed oil was analysised. The main experimental results are as follows:Silica gel column chromatography, Sephadex LH-20 column chromatography of purification experimentation towards alcohol extract components from pomegranate rind. Eventually we got compounds ofⅠ,Ⅱ,Ⅲ, and we also carried out further analysis. From the 13C-NMR of compoundsⅠ, we can see that at d = 99-170ppm has 10 peaks, probably for the carbon skeleton of flavonoids, at d = 19-57ppm has 11 peaks for the two possible monosaccharide or a disaccharide 11 carbon. The concrete structures of compoundsⅠ,Ⅱ,Ⅲhave to be further confirmed.Four reaction systems has been used to determine antioxidant active of pomegranate peel extracts, the results showed that it has strong antioxidant activity. When the concentration is higher than 0.2mg/mL, pomegranate peel extracts has strong reduction than rutin and Vc.The Ec50 of Removal hydroxyl radicals, sodium nitrite, DPPH were 821.97mg/L,528.52mg/L,31mg/L.Pomegranate peel extracts has a certain antibacterial activity, but antibacterial activity of four samples of a certain degree of difference, MIC in the range of 6.25 mg/mL -25.00 mg/mL. In four samples, the ethyl acetate phase has a smaller MIC value,the MIC of E. coli, Staphylococcus aureus, Penicillium, Aspergillus production, reed's yeast, Saccharomyces cerevisiae were 12.50mg/mL,12.50mg/mL,6.25 mg/mL,6.25 mg/mL,6.25 mg/mL,12.50mg/mL。Pomegranate seed oil is abundant in composition, through GC-MS analysis of we know that it containins 10 kind of fatty acids. Higher levels of fatty acids contens palmitic acid (17.87%), linoleic acid (20.05%), oleic acid (21.31%), stearic acid (17.37%),α-linolenic acid (12.82%). Also contains one of the more rare 12.82%α-linolenic acid. In addition, the total fatty acid composition of pomegranate seed oil has three carbon chain fatty acid at carbon 20 and above, Including the squalene (1.33%),which is more rare in vegetable oil. Refractive index of pomegranate seed oil is 1.4898, it means that fatty acid composition of pomegranate seed oil has a relatively large number of double bonds, and with a higher degree of unsaturation. Iodine Value up to 132.926, it shows that pomegranate seed oil is rich in unsaturated fatty acids.Five reaction systems has been used to determine antioxidant active of pomegranate seed oil, the results showed that it has strong antioxidant activity. In the range of 0.2-2mg/mL, pomegranate seed oil has strong reduction than VE. The Ec50 of Removal hydroxyl radicals, sodium nitrite, DPPH were 53.92μL,838.54μL,1.583 mg/mL, the largest clearance rate of inhibiting the activity of phospholipid autoxidation is 47.18%.
