ObjectiveTo investigate the effect of aspirin on proliferation and nuclear factor kappa B expression of Gastric Cancer cell line SGC-7901,AGS and BGC-823.MethodsGastric Cancer cell line SGC-7901,AGS and BGC-823 were treated with aspirin of different concentrations or different time and effect of aspirin on proliferation of SGC-7901,AGS and BGC-823 was evaluated by MTT assay.Apoptosis of SGC-7901,AGS and BGC-823 was detected by DNA agarose gel electrophoresis.Effect of aspirin on cell cycle of SGC-7901,AGS and BGC-823 was analyzed by flow cytometry(FCM). Expression of nuclear factor kappa B p65 in SGC-7901,AGS and BGC-823 was determined by Western blotting and immunofluorescent microscopy.Results(1) Aspirin inhibited the proliferation of Gastric Cancer cell line SGC-7901,AGS and BGC-823.As the incubation time and concentration increasing,the inhibitory effect of aspirin on proliferation was significantly increased.The maximal inhibitory rate occurred at the concentration of 10 mmol/L(P<0.01) and each group had the maximal inhibition after treating for 24 hours.(2) After treating with aspirin of 10 mmol/L for different time, apoptosis was detected by DNA agarose gel electrophoresis in SGC-7901 and AGS and BGC-823.(3) Aspirin mainly blocked the cell cycle in the G0/G1 phase.As the concentration of aspirin increasing,the percentage of cells in the G0/G1 phase of the cell cycle was increased and the percentage of cells in the S phase of the cell cycle was decreased.(4) Aspirin reduced expression of NF-κB p65 in SGC-7901,AGS and BGC-823.Treatments with asprin of 1.0 mmol/L,5.0 mmol/L and 10 mmol/L all significantly inhibited the expression of NF-κB p65(P<0.01). After treating with aspirin of 10mmol/L for 6 hours,12 hours and 24 hours,the expression of NF-κB p65 was significantly reduced(P<0.01).(5) As determined by immunofluorescent microscopy,the expression of NF-κB p65 located in the nuclei of SGC-7901,AGS and BGC-823 cells.Aspirin at 10 mmol/L significantly decreased the expression of NF-κB p65 in cells at 24 hours.ConclusionAspirin inhibits proliferation of Gastric Cancer cell line SGC-7901, AGS and BGC-823.Inhibition of NF-κB p65 expression is one of the mechanisms through which aspirin induces apoptosis of Gastric Cancer cell line SGC-7901,AGS and BGC-823.
