Radix astragali is a common traditional Chinese medicine,usually being known as "the best of all medicine of tonifying Qi".It has decisive position in traditional Chinese medicine and has many curative effects in recent years.Due to the extensive use of Radix astragali and the traditional methods of Radix astragali usage,processing and active components extraction had been unable to meet the needs of people.On the basis of Chinese medicinal herbs preparation,we utilized Lentinus edodes to ferment Radix astragali with the modern bioengineering fermentation technology.The objection of our work is want to achieve its high efficacy and improve the actual utilization The results showed that:Using the single—factor of plate growth experiment and liquid flask shaking experiment,the optimal composition of the culture medium for Radix astragali is:Radix astragali(80eyes),corn flour20g,soybean cake powder 20g,VB150mg,potassium dihydrogen phosphatelg,Magnesium Sulfate 1g,distilled water 1000ml,pH value was natural,inoculum size of Lentinus edodes fermentation broth was 10%.The major active components from fermentation broth were detected after fermented for 1d,4d,8d,12d,15d,respectively.Compared with the control group of Radix astragali,the contents of soluble polysaccharids was decreased,total flavonoids and saponins were increasd from the Radix astragali fermentation supematant.Although they did not reach a significant level,the fermentation duration for Radix astragali should be eight days.In controndition to a preliminary TLC analysis about fermentation broth of Astragalus and a comparison to the spots in the TLC plate that fermentation broth of Astragalus and Astragalus decoction,it showed that fermentation broth of Astragalus had no new components and some components reduced.Some components of Astragalus may be generated into other ingredients by mushrooms fermentation.Comparisons of immunity test results revealed that:In the weight of immune organs and in particular for the impact of spleen weight,Mushrooms blank group of normal mice to promote the role is the best.Effect on the clearance exponent of mouse,fermentation group of Astragalus its adding 100g/L role in promoting is the best.The promotion on the macrophage phagocytosis and the capacity of T lymphocytes from fermentation group that contents of Astragalus was 50g/L was the best.Compared with the control group,experiment group had a certain degree effect of immune enhancing.Detected the positive rate of T-lymphocyte subsets CD4+,CD8+ by the immunocompromised model of mouse further,it revealed that mushrooms broth without Radix astragali and Radix astragali broth without mushrooms were better,but fermentation broth from Radix astragali by mushrooms had no desired results.
