Object:To analyze the variation of cruor function,biochemical indicator,and cytokines of the heat stroke rats,and to determine whether Xuebijing injection can exert protective effects against heat stroke Methods:Adequate anesthesia was maintained by means of a single intraperitoneal dose of 3%Pentobarbital Sodium (1ml/kg).24 SPF bull Sprague-Dawley rats were divided into the following groups:In normal control group with no treatment(n=8),Tco was maintained at about 34℃,at a room temperature of 26℃throughout the entire experiment period.In the Normal saline-treated heat stroke groups,the rats were treated with a dose of isotonic sodium chloride solution via femoral vein(4ml/kg) immediately after the initiation of heat exposure.They were exposed to ambient temperature of 43℃for 47 min to induce heat stroke.Immediately after the onset of heat stroke, the heating pad was removed and the rats were allowed to recover at room temperature(26℃).In the drug-treated heat stroke groups,the rats received an intravenous dose of Xuebijing injection via femoral vein(4ml/kg) immediately after the initiation of heat exposure.They were exposed to ambient temperature of 43℃for 47 min to induce heat stroke.Immediately after the onset of heat stroke, the heating pad was removed and the rats were allowed to recover at room temperature(26℃).Their physiological parameters(MAP,Tco,and HR) were continuously monitored to abserve survival times(intervals between the initiation of heat stroke and the rats' death).Adequate anesthesia was maintained by means of a single intraperitoneal dose of 3%Pentobarbital Sodium(1ml/kg).24 SPF bull Sprague-Dawley rats were divided into the following groups:Normal controls(n=8),Normal saline-treated heat stroke rats(n=8),and Xuebijing injection-treated heat stroke rats(n=8).The methods is the same as above. Fuethermore,2.3ml blood samples at 0,47,and 57 min after the initiation of heat exposure were drawn by arterial femoral cannulation to measure the plasma levels of aPTT,PT,FDP,D-D,Cr,BUN,AST,ALT,ALP,IL-lβ,IL-6,and TNF-α.The rats were killed immediately at 57 min after the initiation of heat exposure to take out of liver which was then fixed by formalin.The paraffin section of liver was dyed by HE.Thepathologic change of liver using light microscope were abserved.Results:Survival time values during heat stroke for rats pretreated with isotonic sodium chloride solution were found to be 15±2.07 min.Pretreatment with Xuebijing injection increased the survival time values to a new level of 74.625±4.627 min(P<0.05).Both two heat-stressed rats display excessive activation of systemic inflammation,hypercoagulation,and tissue ischemia/injury. Excessive activation of systemic inflammation during heat stroke is indicated by increased levels of IL-lβ,IL-6,and TNF-αin plasma.Hypercoagulation is evidenced by increased plasma levels of aPTT,PT,FDP,and D-dimer.Tissue ischemia and injury is evidenced by increased levels of creatinine,BUN,AST, ALT,and ALP in plasma.Xuebijing injection can significantly decrease the plasma levels of aPTT,PT,FDP,D-D,Cr,BUN,AST,ALT,ALP,IL-lβ,IL-6,and TNF-α(P<0.05),and relieve the pathological lesion of liver.Conclusions:Earlier administration of Xuebijing injection before the onset of heat-stress can prolong the survival time of heat stroke rats by decreasing systemic inflammation, hypercoagulation,and tissue ischemia/injury.We can provide valuable time to rescue heat stroke-induced MODS by using Xuebijing injection.
