ObjectiveThe purpose of this study was to explore effects ofelectroacupunture on the structure and function of mitochondria in ischemic penumbra of rats with cerebral infarctionMethodsSprague-Dawley rats were randomly divided into two groups:the sham-operated group and model group.Rats in sham-operated group were only exposed and separated the blood vessel and nerve.The model group was established by the left middle cerebral artery occlusion(MCAO).And the model group was subdivided into two groups:the electroacupucture group,in which the rats were immediately treated for 30 minutes by electroacupumcture after the model was established;and the non-electroacupunture group,in which the left middle cerebral arteries were only blocked.Each group was subdivided into three subgroups at 1 h,3 h and 6 h after operation.Transmission electron microscope was observed by TEM changes of mitochondria structure in the ischemic penumbra.Changes of mitochondrial permeability transition pore were detected by ELITEESP flow cytometer,and Western blotting method was used to detect and analyze the cytochrome c expression in the ischemic penumbra of rats brain;Results1.In the sham-operated group,The mitochondrion shape was nomal,the double membranes were clear.In non-electroacupunture group,neurons were edema, mitochondrion were swelling,its inner and outer membranes were unclear,crista deformed,even broken or disappeared,in electroacupuncture group,mitchndrial structure were better than those of non-electroacupunture group at 3 h and 6 h.but some mitochondia were lighting edema,presenting spherical shape,a few crista breaked or disappeared under transmission electronmicroscope.2.Compared with sham-operated group,the results of FL,FSC,SSC in model group were significantly changed at 1 h,3 h and 6 h(P<0.05),the results of FL,FSC,SSC in electroacupunture group were not significantly different than in the non-alectroacupunture grou(P>0.05).And in sham-operated group,there were no statistical significance between the different times(P>0.05).3.The level of cytochrome c in model groups was increased than in sham-operated groups at 1 h,3 h and 6 h(P<0.05),the expressionl ofcytochrome c in the non-electroacupunture group was also increased than in electroacupunture group at three different time points(P<0.05).ConclusionThe results suggested that electroacupunture may protect neurons through keeping mitochondrial structure and inhibiting cyt c protein released from mitochndria without influencing the MPTP.
