[Objective]To detect the expressional changes of Caspases-1,IL-1βand apoptosis neurocytic in the spinal and cerebral cortex of the adult rats subjected to transected spinal cord injury.[Methods]The model of rats suffered complete cord transaction was used.Terminal deoxynuleotidyl-mediated nick end labeling(TUNEL) was used to detect the expression of neural apoptosis in both caudal and rostral cord stump to the injured site after operation(3,7,14,21 days).The expressions and changes of Caspases-1,IL-1βand apoptosis neurocytic were studied systematically by the methods of immunohistochemical staining in the same model.[Results]1.TUNEL-positive cells stained by yellow and blue can be seen in the whole Spinal cord,positive products located in the nucleus.A great number of TUNEL-positive cells were observed in caudal and rostral cord stump to the injured site at 3 days postoperation,and lasting to 21 days postoperation,presenting a significant increase compared with sham-operated control group(P<0.05),Meanwhile, inner cone cell layer can be seen some apoptosis cone cells.2.There were Caspases-1-positive cells in the sham-operated control group and all operation groups. Compared with the sham-operated control group,the number of Caspases-1-positive cells at 3day and 21day groups increased markedly(P<0.05).In addition,the number of Caspases-1-positive cells in cerebral cortex increased significantly.3.The changes of IL-1βwere similar to the changes of Caspases-1.4.Caspases-1-positive cell can be a IL-1β-positive cell by immunohistochemical staining of the consecutive cells.[Conclusions]1.Apoptosis of neural cells occurred extensively,it may be a main dead pattern in the secondary lesioned period after spinal cord injury.2. Caspases-1-positive cells and IL-1β-positive cells in all operation groups were increased markedly compared with the sham-operated control group.Furthermore. With the immunohistochemicat staining method in the consecutive cells, Caspases-1-positive cell could be a IL-1β-positive cell.According to the results,it is presumed that the Caspases-1 and IL-1βmay be closely associated with the neural apoptosis of the secondary injury following complete transaction.
