ObjectiveDepression is a very serious psychosis disorder in the world nowadays.With the development of the society and the disease spectrum,its influence on people's health becomes distinct day by day.According to the report of WHO, among all kinds of diseases,the financial burden caused due to depression took the fourth place globally.The quota index was expected to jump to the second rank place in 2020.As a result,it becomes urgent for medical experts and scientific workers to search for effective treatment,and clarify its action mechanism.The pathologic mechanism of depression has not been completely revealed scientifically.The monoamine neurotransimtter theory took the leading place in the past.Now general point of view is that psychology is very important to depression.Music therapy and acupuncture are important parts in the therapy of depression.They have a lot of relations,the ancients have recognized this and many studies have proved that in the recent years.Music electro-acupuncture is based on the electro-acupuncture and music therapy, This can alleviate the symptoms of patients suffered from depression to some extent,with its advantage of easy practicing,and with few side-effects,it can adjust psychology and physiology,though the mechanism of its effect remains unclear.Our research focused on observing the effect of Music electro-acupuncture on changes of behavior in depression rats,furthermore, exploring some possible mechanisms by investigating the influence of Music electro-acupuncture on regulating the secretion of monoamine neurotransmitter of rats with depression.Methods:Forty male rats were evenly randomized into control group,model group,music electro- acupuncture group and Fluoxetine group.Depression model was established by using chronic unpredictable mild stress(CUMS) stimulation (forced ice-water swimming,electricity shock,tail-clamping,up-side-down of day and night,fasting,etc) combined with lonely raising for 21 days. Music electro-acupuncture was applied to Baihui(GV 20 ) and Sanyinjiao(SP6),once daily,continuously for 21 days.The rats in Fluoxetine group were stomach filled with Fluoxetine(50mg/Kg/d),once daily, continuously for 21 days.The rat' s body weight,open-field test were recorded on the 1th and 21st day during experiments respectively.After the behavior observation,the contents of monoamine neurotransmitter(NE,DA and 5-HT) in central nerve system of rats with depression were determined.Results:1.Compared with control group,the weight increase was slow and the activities of rats in open-field test in model group were significantly decreased.Music electro-acupuncture can effectively improve behavioral disorder of depression rats.2.Compared with normal group,the levels of Norepinephrine(NE) and Serotonin(5-HT) in prefrontal cortex and hippocampus of model rats with depression were significantly decreased.Music electro-acupuncture can increase the levels of monoamine neurotransmitter in central nervous system.Conclusion1.Music electro-acupuncture at "Baihui(GV 20) and Sanyinjiao(SP6) can prevent or suppress the onset of depression and raise the volume of behavior scores and intake in rats with depression.2.Music electro-acupuncture at "Bai hui"(GV 20) and Sanyinjiao(SP6) can increase the levels of Norepinephrine(NE) and Serotonin(5-HT) in prefrontal cortex and hippocampus of model rats with depression.This might be the one of the action mechanisms of music electro-acupuncture in treating depression. |
