Objective:To observe the influence for protein tyrosine phosphatase-2(Shp-2) of acute leukemia patient marrow original cells with the artesunate(ATS).Methods:Shp-2 was examined by enzyme-linked immunosorbent assay,of leukemia cell line K562 and acute leukemia patient marrow original cells,from exponential growth phase cells,respectively,by adding different concentrations of experimental drugs,divided into:(1) experimental group:①ATS high,mid and low dose group;②ATS high,mid and low dose+transferrin group;③ATS high,mid and low dose+daunorubicin group;④ATS high,mid and low dose+daunorubicin+transferrin group;(2) control group:①medium group;②medium+transferrin group;③medium+daunorubicin group;④medium+ daunorubicin+transferrin group,a total of 16 group.Results:ATS to K562 cells:①Shp-2 of ATS high,mid and low dose group decreased than the medium group(P<0.05),there was no significant difference inter-group of ATS high,mid and low dose groups(P>0.05);②Shp-2 of ATS high,mid and low dose+transferrin group decreased than the medium+ transferrin group(P<0.05),There was no significant difference inter-group of ATS high,mid and low dose+transferrin groups(P>0.05);③Shp-2 of ATS high,mid and low dose+daunorubicin group decreased than the medium+ daunorubicin group(P<0.05),There was no significant difference inter-group of ATS high,mid,low dose+daunorubicin groups(P>0.05);④Shp-2 of ATS high, mid and low dose+daunorubicin+transferrin group decreased than the medium +transferrin+daunorubicin group(P<0.05),There was not statistically significant difference inter-group of ATS high,mid and low dose+transferrin +daunorubicin groups(P>0.05). ATS of acute leukemia in patients with original bone marrow cells:①Shp-2 of ATS high,mid and low dose group decreased than the medium group(P<0.001), Shp-2 level of ATS mid group was lower than ATS high-dose group,and ATS low-dose group(P<0.001);②Shp-2 of ATS high,mid and low dose group decreased than the medium+transferrin group(P<0.001),Shp-2 level of ATS mid dose +transferrin group was lower than ATS high-and low-dose+transferrin group (P<0.001);③Shp-2 of ATS high,mid and low-dose group decreased than the medium+daunorubicin group(P<0.01),Shp-2 level of ATS mid dose+ daunorubicin group was lower than ATS high-and low-dose+daunorubicin group (P<0.001);④Shp-2 of ATS high,mid and low dose+ferritin+daunorubicin group decreased than the medium+transferrin+daunorubicin group(P<0.01),Shp -2 level of ATS mid dose+transferrin+daunorubicin group was lower than ATS highdose+transferrin+daunorubicin group and ATS low dose+transferrin +daunorubicin group(P<0.001).Conclusion:ATS significantly inhibited proliferation of K562 cells, reduced the expression level of endogenous Shp-2 can cause leukemia cell apoptosis and growth inhibition.The strongest role of proliferation inhibition was found to acute leukemia patient marrow original cells in ATS mid-dose group(25μg/ml).Daunorubicin significantly inhibited the proliferation of synergy to acute leukemia patient marrow original cells, jointing ATS mid dose group(25μg/ml).Transferrin was not found to acute leukemia patient marrow original cells in vitro synergy.The role of ATS to acute leukemia patient marrow original cells did not show growth inhibition simple linear dose-effect relationship,there may exist the phenomenon of the best inhibitory concentration.ATS can inhibit Shp-2 protein expression of the acute leukemia patient marrow original cells,thereby to play a role of inhibiting leukemia cell proliferation.
