Objective:A large number of clinical investigations proved that Dang-Gui-Bu-Xue decoction and its ingredients,astragalus(黄芪,Huang Qi) and angelica (当归,Dang Gui),had the function of regulating blood lipids and inhibiting atherosclerosis(AS).This experiment separated and extracted the components of single herb,astragalus and angelica,into astragalus polysaccharide,angelica polysaccharides,angelica volatile oil, astragalus alcohol extract and angelica alcohol extract etc,took Dang-Gui-Bu-Xue decoction as control,and observed the effect of Dang-Gui-Bu-Xue decoction and the extracts of astragalus and angelica on blood lipid and plaque in AS rat model induced by VD3 injection+high-fat diet.Through the horizontal analysis of the pharmacodynamics differences among the drug groups,this investigation explores the concrete effect position and substance basis of Dang-Gui-Bu-Xue decoction to regulate blood lipid and inhibit AS,provided the research base for the further optimization of the compatibility of Dang-Gui-Bu-Xue decoction to regulate blood lipid and inhibit AS,and provided the foundation for the development of new drugs of Dang-Gui-Bu-Xue decoction.Methods:Boiled astragalus and angelica by 5:1 ratio to Dang-Gui-Bu-Xue decoction,then separated and extracted theastragalus and angelica into astragalus polysaccharide,angelica polysaccharides,angelica volatile oil,astragalus alcohol extract and angelica alcohol extract through ethanol-and water-extraction methods. 80 Wistar male rats(200-250g) were randomly divided into 8 groups: control group,model group,astragalus polysaccharide group,angelica polysaccharide group,angelica volatile oil group,astragalus alcohol extract group,angelica alcohol extract group and Dang-Gui-Bu-Xue decoction group.In addition to the control group,rats in the other 7 groups were lavaged with VD3injection and fed with high-fat diet,and rats in control group were lavaged with equal quantity of normal saline and fed with common fodder.After six weeks,under ether anesthesia,sampled from orbital venous plexus for determining blood lipid(TG,TC,HDL-C, LDL-C),calculating the atherogenic index(AI),and detecting whether modeling was success.After the success of modeling,in addition to the control and model groups,rats in every drug group were lavaged with the corresponding drug intervention;the rats in control and model groups were lavaged with normal saline.After four weeks,under ether anesthesia,the rats were sampled from the orbital venous plexus,for determining the blood lipids, calculating the atherogenic index,and analyzing the data in each group with SPSS statistical software.Animals were sacrificed by breaking neck, undertaken the aortic dissection.Then we observed the aorta conditions with light microscopy after HE staining and compared the differences between groups.Results:1.AS rat model was reproduced by VD3 injection+high-fat diet feeding successfully.2.Blood lipid results showed that:TG in astragalus alcohol extract group was siginificantly lower than model group(P<0.05),and TG in angelica volatile oil group higher than model group(P<0.05),suggesting that astragalus alcohol extract could inhibit the increase of TG;TC and LDL-C in astragalus alcohol extract group,angelica alcohol extract group and Dang-Gui-Bu-Xue decoction group was significantly lower than model group (P<0.05),but it showed no significant difference among the three groups (P>0.05),suggesting that astragalus alcohol extracts,angelica alcohol extract and Dang-Gui-Bu-Xue decoction could inhibit the increase of TC and LDL-C,it showed no significant difference among the three groups(P> 0.05),but there existed the trend of efficacy,Dang-Gui-Bu-Xue decoction>astragal side alcohol extract>angelica ethanol extract.TC and LDL-C in angelica volatile oil group increased dramatically compared to the model group and other drug groups(P<0.05),TC and LDL-C in angelica polysaccharides and astragalus polysaccharide groups decreased compared to the model group,but no statistical significance(P>0.05);HDL-C in Angelica volatile oil group increased significantly compared to model group(P<0.05),the remaining HDL-C changes in drug groups were no statistical significance;AI in astragalus alcohol extract and Dang-Gui-Bu-Xue decoction groups decreased compared to the model group, and with statistical significance(P<0.05),the efficacy trend was: Dang-Gui-Bu-Xue decoction>astragalus alcohol extract.The difference of AI in Dang-Gui-Bu-Xue decoction and astragalus alcohol extract groups had a statistical significance(P<0.05),AI in angelica volatile oil group increased significantly compared to model group with a statistically significance(P<0.05).3.After HE staining,aortic under light microscopy showed:in control group,the inner,middle and outer membranes of artery were intact,and with distinctive nuance;there appeared lipid deposition,foam cell formation and calcium salts deposition in model group;there no existed foam cell in Dang-Gui-Bu-Xue decoction group and astragalus alcohol extract group;in angelica polysaccharide group,there were a little of foam cell;in astragalus Polysaccharide Group,there existed proliferation of the inner membranes of artery and lots of foam cells in inner and middle membranes of artery;there were also foam cell in angelica alcohol extract group,but no proliferation in inner membranes; in angelica volatile oil group,there existed fusion of foam cell and coloboma of inner membranes.Conclusions:Astragalus alcohol extract could inhibit the increase of TG,and astragalus alcohol extracts,angelica alcohol extract,Dang-Gui-Bu-Xue decoction could inhibit the increase of TC and LDL-C.And there is a certain efficacy trend,Dang-Gui-Bu-Xue decoction>astragaloside alcohol extract>angelica alcohol extract.Angelica volatile oil regulated blood lipid and the AS negatively;it elevated TC,TG,HDL-C and LDL-C.In addition to angelica volatile oil,Dang-Gui-Bu-Xue decoction and extracts of astragalus and astragaloside also could inhibit plaque.It is speculated that the composition in astragalus alcohol extract and Angelica alcohol extract were the main substance basis in Dang-Gui-Bu-Xue decoction,which could regulate blood lipid;and astragalus polysaccharide,angelica polysaccharides,astragalus alcohol extract and angelica alcohol extract were mainly substance basis which could inhibit AS,but the efficacy of it,as a single medicine efficacy,was not equal to Dang-Gui-Bu-Xue decoction.Maybe because there exist some chemical reaction in the decoction process of astragalus and angelica and with other active ingredients subject to dissolute to enhance the efficacy.
