Objective: To investigate the effect of Chinese herb Dashen-Dahuang on learning and memory ability, cholinesterase (CHE), choline acetyltransterase(ChAT), peroxidase (POD), total antioxidant capacity (T-AOC), the expression of amyloid beta-protein precursor (APP) and presenilin-1 (PS1) in rats with Alzheimer's disease (AD) induced by D-galactose and AlCl3 and its possible mechanisms.Methods: The AD model was produced by injecting D-galactose and AlCl3 intraperitoneally for 90 days. Since the 20th day of D-galactose and AlCl3 intraperitoneal injection, the Dashen-Dahuang group has been treated with Dashen-Dahuang extraction by intragastric administration for 70 days. The control group was produced by injecting the same amount of saline and double distilled water intraperitoneally for 20 days, Since the 20th day, the control group has been treated with saline by intragastric administration for 70 days. Subsequently, ChAT, CHE, POD, T-AOC of the mice's brain tissue was tested by Biochemical methods; learning and memory ability of the mice was evaluated by Morris water maze and hippocampal gene expression of APP and PS1 was tested by reverse transcription polymerase chain reaction (RT-PCR).Results :1. Rats intragastric administration with Danshen-Dahuang, mice had shorter latency (P<0.05) and less error times (P<0.05);2. The Danshen-Dahuang group had obviously decreasing content of CHE and higher content of ChAT than the model group (P<0.05);3. The Danshen-Dahuang group had obviously higher activity of POD and T-AOC than the model group (P<0.05);4. The Danshen-Dahuang group had lower expression of APP and PS1 than the model group (P<0.05).Conclusions: Dashan-Dahuang improve the learning and memory ability of AD rats, its mechanism may be related to the improved acetylcholine system,improved antioxidant capacity,downregulated expression of APP and PS1 mRNA.
