The Study Concerning The Action Of GnRH-a Triptorelinacetate On Human Endometrial Carcinomaline HEC-1B And The Effect On Expression Of P21,PCNA

Objective:By investigate the effects of triptorelinacetate on proliferating inhibition and the effect on expression of P21 and PCNA in endometrial carcinoma cell lines HEC-1B, to explore its role in endometrial cancer mechanism.Methods:â‘ Endometrial carcinoma cell lines HEC-1B was cultured in vitro.â‘¡HEC-1B cells were treated by triptorelinacetate at different concentrations for 24,48,72,96h and the growth curve was drawn.â‘¢HEC-1B cells,treated by triptorelinacetate at different concentrations for 24,48,72,96h were examined for their growth inhibition rates with MTT.â‘£HEC-1B cells were treated by triptorelinacetate in different concentrations(10-9~10-5)for 72h.The expression of P21 and PCNA protein were determined by immunocytochemistry,P21 and PCNAmRNA by RT-PCR.Results:â‘ The result of MTT showed that the auxodrome of human endometrial carcinoma cell lines HEC-1B of triptorelinacetate was S shape.â‘¡The result of MTT showed that triptorelinacetate could effectively inhibit the growth of human endometrial carcinoma cell lines HEC-1B in vitro in concentrations-dependent and time-dependent manners(P<0.05).â‘¢Compared with that of control group,the expression of P21 and P21mRNA,the medication group was significantly increased and PCNA and PCNAmRNA was significantly decreased(P<0.05).Conclusions:Triptorelinacetate can effectively inhibit the growth of human endometrial carcinoma cell lines HEC-1B in vitro in dose-dependent and time-dependent manners.The medication group can enhance the expression of P21 and reduce the expression of PCNA.These show that triptorelinacetate can regulate the expression of P21 and PCNA.The medication group may have some influence on growth inhibition of endometrial carcinoma,involving the levels of protein and gene.
