| Objective: According to the pathogenesis of Alzheimer's disease in both Chinese and western medicine as well as the research progress on Rhodiola herb,we take western medicine Hydergine as the control,by detecting the AD rats'learning and memory abilities through observing their behaviors and by detecting the AD rats's hippocampus tissue's activities of CAT and GSH-PX as well as the protein expression of Caspase-3,to explore the mechanism of Rhodiola herb effect which can improve the AD rats'learning and memory ,to explore the mechanism of Rhodiola herb effect which can treat the AD rats,to provide the experimental basis for clinical medication.Methods:(1)Model:AD model was formed by injecting D-gal(CONC:10mg/ml) into abdominal cavity and taking AlCl3(CONC: 20mg/ml)by intragastric administration as well as injecting scopolamine at last.(2)Grouping:AD models ,which were maken successfully,according to the random number table,were divided into three groups:model group, western medicine (Hydergine) group, traditional Chinese drug(Rhodiola herb) group,setting a blank group.(3)Administration:Blank group and model group rats were intragastrically administered 5ml/kg/d saline to; Rhodiola herb group rats were intragastrically administered Rhodiola herb by a dose of 5g/kg/d; Hydergine group rats were intragastrically administered Hydergine to by a dose of 1mg/kg/d.The intragastric administration lasts for 4 weeks,each medicine's dosage is 20 times more than human being's.All rats were raised under the same condition.(4)Used Morris water maze test devices as well as dynamic image acquisition and analysis system for learning and memory testing, the testing proceedings involved hidden—platform acquisition training and probe trial testing. Various parameters were recorded by computer.(5)Used spectrophotometry to detect the activities of CAT and GSH-PX in the AD rats'hippocampus tissues.(6)Used immunohistochemical detection to analyze the protein expression of Caspase-3 in the AD rats'hippocampus tissues.Results: (1)learning and memory ability Detected by Water maze:Compared with the blank group rats'cognitive abilities,the model group rats'are remarkably decreased(P< 0. 05);The treatment groups rats'cognitive abilities are remarkably improved compared with the model group rats'(P<0.05);There is no remarkably difference between Rhodiola herb group and Hydergine group(P>0.05).(2)CAT detected:The model group rats'activities of CAT in hippocampus tissues are remarkably weakened compared with the blank group rats'(P<0.01).Afeter the treatment ,the treatment groups rats'activities of CAT in hippocampus tissues are remarkably strengthened compared with the model group rats'(P<0.01).(3)GSH-PX detected:The model group rats'activities of GSH-PX in hippocampus tissues are remarkably weakened compared with the blank group rats'(P<0.01).Afeter the treatment ,the treatment groups rats'activities of GSH-PX in hippocampus tissues are remarkably strengthened compared with the model group rats'(P<0.01).(4)Apoptosis-related protein(Caspase-3)detected:The model group rats'protein expression of Caspase-3 in hippocampus tissues are remarkably strengthened compared with the blank group rats'(P<0.01). Afeter the treatment , the treatment groups rats'protein expression of Caspase-3 in hippocampus tissues are remarkably weakened compared with the model group rats'(P<0.05).Conclusions:(1) The AD model ,which was formed by injecting D-gal into abdominal cavity and taking AlCl3 by intragastric administration as well as injecting scopolamine at last,is a better simulation for Alzheimer's disease,is an ideal model for AD.(2)Rhodiola herb can remarkably improve the AD rats'spatial learning and memory impairment , which suggests that Rhodiola herb can prevent and treat AD. (3)Rhodiola herb can remarkably strengthened the AD rats'activities of CAT and GSH-PX in hippocampus tissues,which suggests that eliminating free radicals is one of the mechanisms which Rhodiola herb can improve the AD rats'spatial learning and memory impairment.(4)Rhodiola herb can remarkably weaken the AD rats'protein expression of Caspase-3 in hippocampus tissues,which suggests that Rhodiola herb can protect neurons from apoptosis.There may be relationships between the effect mentioned above and the improvement of AD rat's spatial learning and memory impairment. |
