| Objective To investigate the role of Glial cells of rat affecting the blood-brain barrier and blood tumor barrier P-glycoprotein expression.To investigate The quercetin inhibition effect on the expression of P-glycoprotein in blood brain barrier of glioma model of rat.Methode The c6 Glial cells were cultured in the way of a single adherent cultu regrrowing the Glial cells into the caudate nucleus of rat to made a brain tumor model,blood-brain barrier and blood tumor barrier structure will be observed under microscope after HE staining.Immunization was used to observe the blood-brain barrier and blood tumor barrier endothelial cells P-glycoprotein expression.Result P-glycoprotein expressed in the endothelial cells of the blood-brain barrier and blood tumor barrier,Glioma cells have the expression of P-glycoprotein,too.At the same time the growth of glioma inducted new capillary to formate.Glioma can induce the expression of P-glycoprotein more.tumors in rats of P-glycoprotein-positive rate higher than the sham-operated group,and the tumor group+quercetin group of P-glycoprotein-positive tumors was significantly lower than that group.UV spectrophotometric determination of drug concentrations found that the tumor group+ vincristine+quercetin group vincristine levels in rat brain was more than tumor group+ vincristine group.Conclusion Gliomas can induce P-glycoprotein expressioning much more,quercetin can enhance glioma blood-brain barrier permeability in rats. |
