| Objective:To investigate the correlation between the results of the two methods—FISH and IHC used to test HER2 status for 50 cases of invasive breast cancer.Methods:50 cases of invasive breast cancer were collected randomly from the first hospital of Shanxi Medical University and all of them had accepted operation from 2008 to 2009.We utilized FISH and IHC respectively for processing consecutive cut sheet of each case to test HER2 status,and than analysised the correlation between the results of two methods.Results:1.Seventeen cases(34%)of HER2 gene amplification were detected in 50 eases of breast cancer by FISH.Fifteen cases showed positive expression(3+,2+~3+,2+)based on IHC,and 35 cases were negative(1+,);2,It is proved tha the results of FISH and IHC have correlation by Chi-Square Tests (x~2=23.24,P<0.001);3,Regarding FISH as the gold standard we can get that the sensitivity of IHC is 76.5%,specificity is 93.9%,accuracy is 88.0%,Youden's index is 0.704,positive predictive value is 86.7%and negative predictive value is 88.6%。Conclusion:Since the results of FISH and IHC have excellent correlation,we can regard IHC as screening technique to detect HER2 status for the patients with breast cancer to determin whether Herceptin is choosed.We suggest that the patients with breast cancer proved to be positive(3+,2+~3+,2+)based on IHC should choose FISH to diagnose. |
