EGFR MRNA And Angiogenesis, Chemotherapy Resistance In Ovarian Carcinoma

Objective To investigated the expression and significance of the EGFR mRNA,vascular endothelial growth factor(VEGF) mRNA and lung resistance protein(LRP) mRNA in ovarian carcinoma.Methods Reverse transcription polymerase chain reaction(RT-PCR) was employed to determine the expression of EGFR,VEGF and LRP mRNA in 79 ovarian specimens(including 15 normal,13 benign and 51 malignant),and their relationship was analyzed.The chemotherapic effect after operation was followed up.Results(1)The positive rates of EGFR,VEGF and LRP mRNA were significantly higher in patients with ovarian carcinoma than those in ovarian benign tumors and normal controls (P＜0.05).(2)There was correlation between EGFR mRNA expression and clinical stage,stageⅢ-Ⅳhad higher positive expression rate and expression level than stageⅠ-Ⅱ(P＜0.05).The expression of VEGF mRNA was correlated with the clinical stage and lymph node metastasis. VEGF mRNA expression level in stageⅢ-Ⅳand in the group with lymph node metastasis were significantly higher than those in stageⅠ-Ⅱand in the group without lymph node metastasis respectively(P＜0.05).The expression of LRP mRNA was not associated with the clinicopathologic parameters in epithelial ovarian cancer(P＞0.05).(3) Analyzing follow-up data,EGFR,VEGF and LRP mRNA expression level in patients with chemotherapy sensitive were lower than those in the resistance ones(P＜0.05).(4) The expression of EGFR was positively correlated with the expression of VEGF(r=0.460,P＜0.05) and LRP(r=0.749,P＜0.01).Conclusions(1) The expressions of EGFR and VEGF mRNA are positively correlated to the occurrence of ovarian carcinoma and its metastasis.(2) The expression of EGFR in ovarian cancer was related to angiogenesis.(3)The high level of EGFR,VEGF and LRP mRNA expression were related to chemotherapy resistance,detecting them may be helpful in monitoring chemotherapy resistance for ovarian cancer.
