Objective:(1) To investigate the expression of OPN,S100A4 in breast tissue and examine the correlations with clinicopathological parameters.(2) To investigate the serum concentration of OPN in breast tissue and examine the correlations with clinicopathological parameters.Methods: We investigated the protein expression of OPN and S100A4 in 86 cases of breast carcinomas ,48cases of benign lesions and 30 cases of control of the tumor adjacent to normal breast tissue using immunohistochemistry.The preoperative(86 cases breast carcinomas , 48 cases of benign lesions ,48 cases of physical examination )and postoperative(86 cases breast carcinomas, 19cases metastasis and 67 cases without metastasis )serum concentrations of OPN was measured by enzyme-linked immunosorbent assay. Data obtained from the results of the application package SPSS13.0 statistical analysis to P <0.05 for the difference has statistical significance.Results:(1) The positive expression rate of OPN was 73.26%(63/86),31.25﹪(15/48) and 20.0﹪(6/30)in tumor tissue,benign lesions and the tumor adjacent to normal breast tissue ,respectively.The positive expression rate of OPN was elevated significantly in tumor tissue compared to benign lesions(P<0.05).The positive expression rate of S100A4 was 63.95%(55/86),20.83﹪(10/48) and 0.00﹪(0/30)in tumor tissue,benign lesions and the tumor adjacent to normal breast tissue ,respectively.The positive expression rate of S100A4 was elevated significantly in tumor tissue compared to benign lesion(P<0.05). (2)There was significant higher expression of OPN in lymph node involvement extrathyroidal invasion, TNM stages,C-erbB-2 (P<0.05),there was no significant difference among the expression of OPN in age,large tumor size,Menstrual status,the degree of tissue differentiation,Pathological types,ER,PR and P53 (P>0.05). Spearman correlation analysis of OPN and the number of axillary lymph node metastases, TNM stages ,C-erbB-2 have positive correlation (P <0.05), r = 0.291,0.254,0.237, respectively. (3) There was signify -cant higher expression of S100A4 in lymph node involvement invasion and P53 (P<0.05),there was no significant difference among the expression of S100A4 in age,large tumor size,Menstrual status,the degree of tissue differentiation,Pathological types,TNM stages,C-erbB-2,ER and PR (P>0.05). Spearman correlation analysis of S100A4 and the number of axillary lymph node metastases and P53 correlation, r = 0.367,. r = -0.391(P <0.05) respectively. (4) The relationship between OPN protein expression and S100A4 protein expression has the correlation r = 0.597, P<0.05 .(5)The preoperative serum levels of OPN was elevated significantly in breast carcinomas patients when compared to the benign controls (P<0.05).there was no signify- -cant difference between the preoperative serum levels of benign lesions and control of physical examination (P>0.05).(6)The preoperative serum level of OPN was elevated significantly when compared to the postoperative control. there were no significant differences among the serum levels of OPN Menstrual status,the degree of tissue differentiation,age (P>0.05) .Breast carcinomas tissue OPN protein expression and serum levels of OPN has relevance (P <0.05), and significantly positive correlation, r=0.361. Conclusions:(1)OPN and S100A4 were over expressed in the breast carcinomas compared to benign tissues. OPN and S100A4 in breast carcinomas tissues of high expression levels can be used as the clinical diagnosis of breast carcinomas support indicators, and as a differential diagnosis of benign and malignant breast indicators .(2) To detect OPN and S100A4 in breast carcinomas expression levels, we get the message breast carcinomas to the clinical situation, but also for breast carcinomas diagnosis, differential diagnosis and formulate a reasonable treatment plan, assessment and prognosis of metastasis and provide a theoretical basis. (3) OPN levels in peripheral blood can be used as indicators of breast carcinomas diagnosis and,it can be used benign and malignant breast disease differential diagnosis of breast carcinomas can also be used to review the monitoring indicators of relapse, as well as clinical assessment of tumor invasion, metastasis, prognosis, etc. (4)OPN in peripheral blood of breast cancer detection can be used as a useful indicator of non-invasive screening.
