Objective CTLA-4 plays an important inhibitory effect on normal T lymphocyte proliferation and activation. It is key to maintain lymphocyte homeostasis in the body. But it prompts CTLA-4 gene polymorphism may result in the autoimmune diseases, and type 1 diabetes are closely related. CTLA-4 carrying inhibitory signals also plays an important role in the transmission of the immune tolerance induction. CTLA-4 gene polymorphism (AT) n repeat of T1DM recessive gene may control the stability of mRNA. The non-coding region in the 3 'region of CTLA-4 is relevance with T1DM susceptibility . In this paper, the study was untaken to determin CTLA-4 gene polymorphism associated with type 1 diabetes in Guangxi Zhuang, Han populations, and learned about the impact of CTLA-4 gene polymorphism on susceptibility to T1DM.Methods A total of 99 subjects of Zhuang, Han populations and 105 participants of Zhuang,Han Chinese as control group were surveyed. Gene polymorphism was determined by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) combined with gel electrophoresis, and then confirmed by direct sequencing. The results were compared with those in 105 matched subjects of Han Chinese from the same region.Results (1) Guangxi Han population CTLA-4 gene microsatellite polymorphism has 15 alleles, Zhuang crowd CTLA-4 gene microsatellite polymorphism with 12 alleles, gene frequency of the two groups showed no statistical significance (P>0.05) . (2) 106bp allele in Han T1DM group was significantly higher than that in the normal control group of Han populations (P = 0.01); 124bp allele frequency was significantly higher in the Han normal control group than in the Han T1DM group (P = 0.01). (3) In the Zhuang T1DM group 88bp allele is higher than in the normal control group (P = 0.01). However, in Zhuang normal control group 124bp gene frequency higher than in the Zhuang T1DM group (P﹤0.05)Conclusion (1) CTLA-4 (AT)n gene microsatellite polymorphism was strongly associated with T1DM patients of Han and Zhuang nationality in Guangxi province. (2)CTLA-4 (AT)n 106bp may be the susceptible gene in T1DM patients of Han nationality in Guangxi. (3)CTLA-4 (AT)n 124bp may be the protective gene in T1DM patients of Han nationality. (4) CTLA-4 (AT)n 88bp may be the susceptible gene in T1DM patients of Zhuang nationality.
