Rosemary is a perfume plant that originally from mediterranean area,and is now world wide planted,also in china recent years.Ever since fifties last century,it is found that rosemary is not only an important perfume,many of its active ingredient have strong characteristics of antioxidant,and most of which is of high medicinal value.The rosemary oil is the volatile component of rosemary,and the most important of which is terpenoid,including monoterpene,sesquiterpene etc.Now,the rosemary oil widely applicated in medicine,daily medical industry,food industry and so on,it is considered a bigger application in the future.Firstly,method of producing rosemary essential oil nanoemulsion and detection of essential oil scontent of rosemary essential oil nanoemulsion is studied,including screening surfactants, plotting pseudo-three-phase plan and HPLC detection method.Secondly,quality evaluation is proceed on rosemary essential oil nanoemulsion,including physics and chemistry,diameter detection,stability examination.Finally,in mice plasma,heart,liver,spleen,lung and kidney for the various biochemical indicators of antioxidant study compared the past and rosemary essential oil and rosemary oil nanoemulsion antioxidant activity in different areas.The content and conclusion is as follows:1 Cofirm the formula of nano-emulsions by selecting from different kinds of surface active agent and mixed surface active agent which contains different HLB,the paper confirm the surface active agent of essential oil of rosemary.It is the mixed emulsifiers of nonionic surfactant Tween-80 and Span-80,in which HLB is 12.Also by analysis the pseudo-ternary phase diagrams we can confirm the area in which can form rosemary essential oil nanoemulsion:the emulsifying agent/rosemary essential oils/water system contains 1% emulsifying agent and 10%rosemary oil.The largest water inflow of this system is 89%.2 Identified nanoemulsion rosemary essential oil essential oil content in the HPLC method: determination of the characteristics of components 1,8-cineole of essential oils,calculated the content of essential oils by content of characteristic components.Chromatographic conditions:BDS column(4.6mm×25cm,5μm);mobile phase:methanol:water(83:17);flow: 1ml/min;detection wavelength 266nm;injection volume 20μl;column temperature 35℃.Of 1,8-cineole standard for precision experiments,RSD=0.90%o Of 1,8-cineole stability standard experimental,RSD=0.28%。Of 1,8-cineole standard to reproduce the experiment, RSD=0.33%。3 The appearance of rosemary essential oil nanoemulsion is white,transparent,and with a certain degree of mobility.Through the methylene blue and sudanⅣtest combined with the proliferation of the relevant parameters to determine the preparation of the rosemary essential oil nanoemulsion are O/W nano-emulsion.Viscosity 0.93s,pH value 7.68,conductivity 24.3μs/cm。In the transmission electron microscope observation of the vast majority of spherical,well dispersed,with an average particle size of about 180nm.Potential particle size analyzer measured the size of about 178nm.The two are basically the same results.4 In the study of the parameters stability of nano-emulsion,the stability of the test parameters to speed up the experiment,light to speed up laboratory tests show that the content and appearance of nano-emulsion have no significant changes in traits.Rosemary essential oil nanoemulsion in aqueous solution was significantly higher than that of the sustained-release properties of rosemary oil in aqueous solution of the release,the results indicate that rosemary oil was after surfactant package to enhance its effective lifetime.The methods established by using HPLC,can accurately calculate the nano-emulsion of rosemary essential oil of rosemary essential oil content of 25.89%.5 By analysis the antioxidant enzymes of the rats' activities of SOD,CAT,GSH-PX and contents of lipid peroxides MDA in the positive control group,rosemary essential oils group, rosemary essential oil nanoemulsion Group,the paper do some research on antioxidant activity which influenced by rosemary essential oils or rosemary essential oil nanoemulsion.The conclusion is as below:rosemary essential oil nanoemulsion not only can do a dose-dependence increasing on the SOD activities and CAT activities to the cardiac tissues of rats,but also dose-dependence increase the tissue of liver activities of GSH-PX enzymes.It also can make a dose-dependence decrease on the MDA of tissue of liver and lung.All the above have improved that rosemary essential oil nanoemulsion could increase the antioxidant enzyme activity and decrease the effect of lipid peroxides in rats.The effect is better than rosemary essential oils.
