Objective:In this study,we want to investigate the association of PC-1 K121Q polymorphisms with GDM in Han nationality women . To assure whether the genes are susceptible or protective gene to GDM.Meanwile,we study the PC-1 K121Q polymorphism of the two newborns.We also want to investigate the association of PC-1 K121Q polymorphisms and IR. We study the differences of Glycosylated hemoglobin and C-peptide between two groups to revel the value of these two clinical data.Methods:100 unrelated subjects were divided into two groups,women with GDM and pregnant women with normal glucose tolerance(n=50 for each group).Cases would be excluded if some conditions occure such as hypothyroidism , hypertension , which may interfere with insulin sensitivity.Two groups were similar in age,parities and gestational weeks.The genotypes of PC-1 K121Q were determined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)assay in two groups.The clinical data were also analyzed.Results:1.The PC-1 121Q allele frequency was higher in women with GDM compared with control group(32%vs8%,P<0.05),the genotype frequency was also higher in GDM than control group(19%vs4%,P<0.05).2.The fasting plasma glucose,large infant subjects,C-peptide,Glycosylated hemoglobin,fasting serum insulin,insulin resistance index were higher in women with GDM compared with control group(P<0.05)3.In GDM group ,PC-1 121Q allele-carrying subjects showed higher level of fasting serum insulin,C-peptide,insulin resistance index(P<0.05)compared with K121K individuals.Conclusions:1.The PC-1 K121Q polymorphisms exist in pregnant women(most are K121K polymorphisms).2.PC-1 K121Q variant correlates with the pathogenesis of GDM,suggesting that PC-1 121Q allele is a susceptible gene to GDM.3.In GDM patients,the PC-1 K121Q variant has independent and adverse effect on causing IR. It could be used to guide the prevention of GDM and to predict the severity level of GDM.4. C-peptide and Glycosylated hemoglobin were different in two groups. In GDM group,PC-1 121Q allele-carrying subjects showed higher level of C-peptide. So,C-peptide and Glycosylated hemoglobin can be used to predict the severity level of GDM.
