Objective:To provide experimental evidence for the estalishment of quality standards of the Traditional Mongolian medicine 'Loudoucai'.Methods:1.The textual research of herbalism and pharmacognostical study on 'Loudoucai'.2.The microscopical identification,physichemical identification and thin layer chromatographic(TLC) methods were used for the qualitative analysis.3.The content of luteolin and caffeic acid in Aquilegia Viridiflora pall.were measured by HPLC method and the content of amino acid was measured by amino acid analyser.4. The content of moisture,total ash,acid-insoluble ash and extract were also measured. Results:The textual research of herbalism and pharmacognostical study on 'Loudoucai' showed that the traditional Mongolian medicine 'Loudoucai' was the dry herb of the perennial plant Aguilegia viridifloa Pall.The microscopic characteristics of the roots,stems,leaves,and powder of Aquilegia Viridiflora pall were detected by microscopical identification;The existence of alkaloids and flavonoids in the herb was detected by physichemical identification;The content of berberine,luteolin,and caffeic acid were detedcted by TLC method;N-butanol - acetic acid - water(7:1:2), benzene - ethyl acetate - isopropyl alcohol - methanol - water(6:3:1.5:1.5:0.3),ethyl acetate - butanone - methanol - water(10:6:1:1) were used as mobile phase to detect berberine under 365nm wavelength.Cyclohexane-ethyl acetate-formic acid(10:10:2), n-hexane-ethyl acetate-formic acid(10:8:2),chloroform-methanol-formic acid(7:2: 0.5) were used as mobile phase,1%ethanol solution of aluminum chloride was used as color development reagent to detect luteoli.Benzene-chloroform-glacial acetic acid (6:1:0.5),chloroform-methanol-formic acid(7:1:0.1),benzene-ethyl acetate-formic acid(3:1.5:0.3) were used as mobile phase to detected caffeic acid.The chromatographic conditions for determination of luteolin: Hypersil-C18ODS(150mm×4.6mm,5μm) column;Mobile phase:methanol-0.4% phosphoric acid(55:45);flow rate:0.6ml/min;wavelength for detection:350nm; column temperature:25℃.The results showed that the content range of luteolin was 0.006875~0.034375μg with a good linear relationship with peak area(r = 0.9992); the average recovery was 95.92%,RSD was 1.95%(n = 5).The chromatographic conditions for determination of caffeic acid:ODS(150mm×4.6mm,5μm,number: E1718531);mobile phase:methanol -0.05%phosphoric acid(17:83.);flow rate: 1ml/min;wavelength for detection:323nm;column temperature:25℃.The results showed that the content range of caffeic acid was 0.0076~0.038μg with a good linear relationship with peak area(r = 0.9995);the average recovery was 96.83%, RSD was 2.00%(n = 5).In total 17 amino acids were detedted from Aquilegia Viridiflora pall.and total content of them was 4.808%,including 7 essential amino acids in the human body(total content of them was 1.689%,35.13%of total amino acids).The content of moisture,total ash,acid-insoluble ash and extract were measured as 9.57%,9.62%,0.94%and 25.2%,respectively in the herb.Conclusions: The result of textual research of herbalism and pharmacognostical was accurate.The microscopic characteristics were clear;physichemical identification method was simple and sensitive;TLC method was specific,reproducible;HPLC method was sensitive,specific,reproducible and its detecting results was reliable.
