ObjectiveNew materials applying to human body must have full-scale biology detection, now analyzing dental alloys biocompatibility by apoptosis correlation factor caspase-3 which assisted by time gradient,and make the alloys applied to clinical use to develop native product.Try to add new method to oral materials biocompatibility test on molecular level,from apoptosis way.Approach the correlation between caspase-3 expression and oral materials biocompatibility,and making it to be one of the factors of evaluating oral materials biocompatibility.Materials and Methods1.Cells cultureL929 cells were cultured with RPMI 1640 cell medium containing 10%fetal calf serum in the incubator of 37℃and 5%CO2.2.Observation of cell morphologyEstablished negative control,Ni-Cr alloys,Ag-Pd alloys,Co-Cr alloys,2# alloys and 3# alloys served as control.Cultivated L929 cells by 6 kinds of liquor for 48 hours, observed living cells,apoptosis cells and dead cells by fluorescence microscope.3.Detection of caspase-3 by absorption spectrometry.Established negative control,Ni-Cr alloys,Ag-Pd alloys,Co-Cr alloys,2# alloys and 3# alloys served as control.Cultivated L929 cells by 6 kinds of liquor for 4 hours,7 hours,24 hours and 48 hours,then collected cells,extracted proteinum,after dyeing, detected OD by Absorbance Microplate Reader,calculated the activity of caspase-3.Results1.The cells' appearance of each group after cultivated 48 hours by leaching liquor.Observed by fluorescence microscope,the cell quantity of each group was different,there were lots of apoptosis cells in each group,and orange cells which were dead or in advanced stage of apoptosis could be seen in Ni-Cr alloys group.2.The activity of caspase-3 in each group with 6 kinds of liquor.The activity of caspase-3 in each group was different(P<0.05).From high to low was:Co-Cr alloys group>2# alloys group>3# alloys group>Ag-Pd alloys group>Ni-Cr alloys group.3.The activity of caspase-3 at different time.The activity of caspase-3 on the spot of 4 hour,7 hour,24 hour and 48 hour was obviously different(P<0.01).From high to low was:4 hour,48 hour,7hour,24 hour.Conclusions1.Each group biocompatibility from good to bad was:Co-Cr alloys group and 2#alloys group>3# alloys group>Ag-Pd alloys group>Ni-Cr alloys group.2.Both the 2#alloys and the 3# alloys may be considered to be applied to clinic.3.The correlation factor of apoptosis,caspase-3,may be possible served as a reference index in evaluating metal materials biocompatibility which should be assisted by time gradient.
