ObjectiveTo study the expression of osteopontin protein(OPN) in the mRNA and protein level of multiple sclerosis(MS) patients.So that to explore the relationship between OPN and multiple sclerosis,and investigate the role of OPN in disease activity with multiple sclerosis and its clinical significance.Methods16 blood samples of MS patients(11 females and 5 males,mean age 34.6±11.5 years,range 19-52 years)were collected from the Department of Neurology,Shengjing Hospital of China Medical University from June in year 2007 to January in year 2009.The mean interval from the diagnosis of MS to the time when the patients were evaluated for this study was 1 month to 3 years.Diagnosis of MS was established according to the diagnosis standard of poster in 1983.20 sex- and age-matched healthy volunteers(16 females and 4 males,mean age 33±7.3 yeas,range 18-51 years)were recruited from Medical Examination Center,Shengjing Hospital of China Medical University as controls.16 blood samples of MS patients were collected again after the 2 weeks.Detection the expression of mRNA level that OPN gene in peripheral blood mononuclearcell(PBMC) with 16 MS cases and 20 normal cases by us(?)ng RT-PCR method and investigation the proportion that CD4~+T cell of osteopcntin signed account of by low cytometry.The data was analyzed by SPSS13.0.Results(1) The method of RT-PCR detected that the expression index of mRNA on MS group and control group seperately was 0.5947±0.2772 and 0.2513±0.1389,and the difference is significant by statistical two samples t test(t=7.922,P<0.01).(2) The result of flow cytometry displayed the positive percentage of OPN in MS group(that is 23.5%) had higher than normal group,that is 16.30%(P<0.01).(3).OPN of MS post-treatment group(that is 17.4%) had lower than MS pretherapy group,that is 23.5%.Conclusions1.Osteopontin may play a important role on the onset of MS.And it also has a promotion on the onset of MS.2.OPN gene of MS group in mRNA has higher express.OPN can be regarded as markers for early diagnosis of MS.
