BackgroundLumbar-disc degeneration(LDD)is a major cause of low back pain. Approximately 80%of the global population experience low back pain in their life time, presenting a huge medical and economical burden to society。Intervertebral disc degeneration(IVDD)associated with spinal degeneration disease,which include disc herniation,radiculopathy,myelopathy,spinal stenosis,ins-tability,and low back pain, constitude the vast majority of the diagnoses treated by spine specialists.the most common form is protrusion of intervertebral disc.Asporin originally was characterized as a member of the class I small leucine-rich repeat pro-teoglycan(SLRP)protein family,which is a subgroup of extracellular matrix proteins in the leucinerich repeat(LRR)protein superfamily.Human asporin is about 50%identical(and 70%similar)to decorin and biglycan(BGN).recently study found asporin significant increase in articular cartilage of the OA patients,so asporin is associated with OA.Given that OA and LDD are both degenerative diseases of skeletal joint regions, and that many of the genes expressed in cartilage are also expressed in the intervertebral disc,we hypothesize that ASPN is an excellent candidate as a susceptibility gene to LDD.My objective is studying the association between asporin and LDD.PurposeTo study the expression and clinical significance of asporin in human degenetation disc.MethodsTotal protein sample used in this study were isolated from 25 patients with protrusion of intervertebral disc who underwent operation at the First Affiliated Hospital of China Medical University.besides,we clloected 5 cases of normal intervertebral disc.these tissue samples were obtained after anterior operation because of lumbar fracture. Western blot was used to detect the expression of asporin in 25 patients with protrusion of intervertebral disc,10 patients with lumbar fracture after anterior operation. we scale gray level for data analysis using ScnImge sdftware and statistics.two simple sample t-test was used to evalue statistical significance.differences were considered as significance when p values were less than 0.05(P<0.05).Results1,The expression of collagenⅡprotein was significance different between normal discs and degeneration discs;the expression of collagenⅡprotein was not significance different between degenetation discs.2,The expression of aggrecan protein was not significance different between normal discs and protrusion discs;the expression of aggrecan protein was significance different between normal discs and extrusion discs,and the expression between degeneration discs was significance different.3,The expression of asporin protein was significance different between normal discs and degeneration discs;the expression of asporin protein was not significance different between degenetation discs.Conclusion1,Expression of asporin are both in noemal discs and degeneration discs.2,Expression of asporin is significant increase associated with the degree of disc degeneration.
