The Impact Of Changyuning Granule To Ulcerative Colitis Rats On IL-1 And Related Clinical Studies

Objective:Through the establishment of rat model of ulcerative colitis to study the influence of the changyuning on the expression of IL-1 which is in rat ulcerative colitis tissue.To investigate the mechanism of Chinese medicine of the treatment of ulcerative colitis on levels of cytokines.To study the clinical efficacy of changyuning through clinical trialsMethods:1.ExperimentalStudy:Bypluschemical-inducedimmune-stimulat ing complexes mathod,150 purebred wistar rats was manufacturing into a rat model of ulcerative colitis.Randomly divided into 5 groups:control group, model group,group of western medicine,Chinese medicine low dose group, high dose group of Chinese medicine.Treatment for 4 weeks,body weight,histopathological changes,Color in rats were observed and score.To measure the levels of IL-1 of rat colon tissue by ELISA,and analyzed statistically.2.Clinical Research:The 30 patients who were diagnosis as cases of ulcerative colitis were randomly divided into treatment group and control group.The patients of treatment group were treated by Changyuning,and The patients of control group were treated by SASP,the safety and efficacy of Changyuning were observed.Results:1.The experimental results:after model,the stool of the rat become thin,some of the stool is mucopurulent bloody stool,some rats lose weight and reduce activities.Histopathology of ulcerative colitis mucosa showed a notable feature which can be seen in microscope,congestion and edema,inflammatory cell infiltration,the goblet cells reducing,crypt abscess.After treatment by Chinese medicine,these symptoms are all improved.It is shows that the treatment effectiveness of high dose group of Chinese medicine is the best,and the treatment effectiveness of group of western medicine is same to the treatment effectiveness Chinese medicine low dose group.the extent of ulcers and erosion in the rat colonic mucosa are all relieving.Epithelial repairing and granulation tissue hyperplasia in the rat colonic mucosa can be seen in the microscope.There are still some lymphocytic infiltration.The levels of IL-1 expression in colon tissue of model group rats is increasing,but that is decreasing in colon tissue of group of western medicine,Chinese medicine low dose group and Chinese medicine high dose group.It is shows that the decrease of high dose group of Chinese medicine is the largest,and the treatment effectiveness of group of western medicine is same to the treatment effectiveness Chinese medicine low dose group.2.The clinical results:The total effective rate in treatment group is.The total effective rate in Control group is.Comparing the two groups,with statistically significant differences(P<0.05).Treatment group is better than the control group in improving symptoms in patients,Comparing the two groups,with statistically significant differences(P<0.05).Conclusion:1.Chang-yu-ning granule may has curative function on rat model of ulcerative colitis,and the high dose group has more Obvious effect.2.Chang-yu-ning granule may has curative function on rat model of ulcerative colitis,the DAI score of the rat model have a very good improvement.3.Chang-yu-ning granule on rat colonic mucosa of IL-1 levels may have reduced the role.4.Two groups of patients with intestinal lesions in varying degrees to imp rove the treatment group than the control group,It is statistical significance...
