| [Object]:By observing the effect of electro-acupuncture on the behavior PARP1 after acute spinal cord injury in rats,try to show that the mechanisms of treatment of electro-acupuncture to ASCI through reducing the expression of PARP1.[Methods]:Two hundred and twenty healthy clean female Wistar rats were randomly divided into electro-acupuncture group,Z-DEVD-FMK group,model group and sham-operation group.There are eleven rats in every group.And every group was divided into 6h,ld,3d,7d,14d time points,eleven rats in every point.With improved Allen's method to make rats acute SCI.The neurofunctional comeback of acute spinal cord injury was measured by BBB score experiment.The segments of injured spinal cord were cut for morphological studies after injury by HE staining.The expression of PARPI of the segments of injured spinal cord were detected by immunohisto chemical staining.[Results]:First:The result of BBB score experiment after ASCI in rats shows:scores of the model group and sham-operation group did not change.The grade of neurofunction in electro-acupuncture group was more improved distinctly than in Z-DEVD-FMK group(P<0.05).Death of rats in electro-acupuncture group is less than in Z-DEVD-FMK group.Second:The effect of expression of electro-acupuncture on PARP-1,PARP-1 splitting fragment after spinal cord injury in rats.(1)PARP1:in the sham-operated group the expression of positive cells.In the model group,the beginning of the expression of 6h after injury,the highest 1d after injury,3d,7d started to decline,14d minimize expression, and in the 1d,3d model group was higher than the expression of sham-operated group(P<0.05).In 1d,3d,the expression of the drug group is less than the model group(P<0.05).In 3d,the expression of electro-acupuncture group is less than the model group(P<0.05).It can be seen:the expression of EA group was higher than the drug group,drug group effect can be seen better than electro-acupuncture group.(2)PARP-1 splitting fragment:In the sham-operated group the expression of positive cells.In the model group,started 6 h after injury the expression of 1d after injury,the highest expression levels in the 3d,7d,14d began to decline,and in the expression of 1d model group was higher than sham operation group(P<0.05).In 1d,3d,the expression of the drug group is less than the model group(P<0.05).In 1d,14d,the expression of electro-acupuncture group is less than the model group(P<0.05).It can be seen:electro-acupuncture group in 6h,1d,7d,14d expression is less than the drug group,one can see the effect of electro-acupuncture group was better than the drug group.[Conclusion]:First:Electro-acupuncture can significantly promote the recovery of hindlimb motor function of ASCI rats.Second:Electro-acupuncture can significantly reduce the expression of PARP1 and PARP1 splitting fragment in the part of spinal cord injury in ASCI rats,show that electro-acupuncture play an important role in the treatment of ASCI rats. |
