Objective:To type 48 SNP loci in X chromosome by SNPlex platform and evaluate their application values for kinship testing.Methods:A 48 X-SNP pool selected from 167 X-SNP loci by Bioinformatics was formed.272 blood samples were genotyped by SNPlex technique.The sensitivity of the method was detected and the genotyping results were verified by sequencing.The forensic application values of 48 X-SNP loci were evaluated through genetic polymorphic investigation.Results:Allele frequencies and genetic polymorphic parameters about 48 X-SNP loci were got.The coincidence rate of of genotying and sequencing was 99.17%. Statistical analysis about heterozygosity,polymorphism information content,the probability of discrimination power and the probability of exclusion showed that the 48 X-SNP loci were highly polymorphic except rs6527549.Conclusion:The X-SNP loci were polymorphic and suitable for forensic biology application.Being highly specific,highly accurate,good compatible,middle-throughout on loci and high-throughout on samples,SNPlex technique meet the needs of forensic biological detection.
