| objective:To observe the changes of macrophages in adipose tissue of insulin resistance(IR) rats on high trans fatty acids diet and saturated fatty acids diet,and explore the relation between IR,obesity and macrophage in adipose tissue.Methods:30 Sprague-Dawlay(SD) rats were divided into three groups randomly,ten rats for each group.Group NOR(normal control group) rats were fed with general diet.While group SFA(saturated fatty acids diet group)rats were fed with high saturated fatty acids diet.Group SFA (trans fatty acids diet group) rats were fed with high trans fatty acids diet.The experiment lasted 8 weeks.At the end of 8 weeks,the rats w hose fasting blood glucose(FBG) and fasting insulin were tested in order to be sure insulin resistance(IR) rat model was established successfully.Then all rats were killed.Taking the samples,body weight(BW), fasting blood glucose(FBG),fasting insulin(FINS),serum triglyceride(rG) and total cholesterol(TC),high density lipoprotein cbolesterol(HDh-C),low density lipoprotein cholesterol(LDL-C),free fatty acids(FFA),visceral adipose tissues weight(VW),in different group rats were measured.To calculate insulin resistance index(IRI) and VW/BW.Histological and immunohistochemical analyses of epididymis adipose tissues were performed for mean adipose cell diameter and CD68 protein expressing.All data were analyzed using SPSS11.0 for windows. Results,Fasting blood glucose(FBG):there was no significent difference (all P>0.05)between the three groups.The serum LDL TG TC LDL/HDL FFA of group TFA and group SFA were higher than group NOR(P<0.01 or P<0.05), And the serum HDL of group TFA was lower than group NOR and group TFA (P<0.01),LDL/HDL of group TFA was higher than group NOR and group SFA (P<0.01).Fasting insulin(FINS):group TFA and group SFA were higher than group NOR(P<0.05),there was no significent difference between group TFA and group SFA(P>0.05).Insulin resistance index(IRI): group TFA and group SFA were higher than group NOR(P<0.05),there was no significint difference between group TFA and group SFA(P>0.05).Body weight(BW):group SFA were higher than group NOR and group TFA(all P<0.01), there was no significint difference between group TFA and group NOR (P>0.05).Visceral adipose tissues weight(VW):group SFA were higher than group NOR and group TFA(all P<0.01),group TFA was higher than group NOR(P<0.01).VW/BW:group TFA and group SFA were higher than group NOR (P<0.01).Mean adipose cell diameter of epididymis adipose tissues: group TFA and group SFAwere higher than group NOR(P<0.01).CD68 protein expressing of epididymis adipose tissues:group TFA and group SFA were higher than group NOR(P<0.01).Dependence Analysis,CD68 protein expressing of epididymis adipose tissues was significantly and positively correlated with diameter of the epididymis adipose cell(r=0.702,P<0.01),VW/BW(r=0.777,P<0.01),and IRI(r=0.804, P<0.01).Conclusion,1,High trans fatty acids diet could induce insulin resistance rat model successfully.2,Serum TC,LDL,TG,LDL/HDL,FFA were markedly increased by high saturated fatty acids diet or high trans fatty acids diet,while HDL was significantly lowered after high trans fatty acids diet.3,Viseral adipose tissue weight,VW/BW,mean diameter of the epididymis adipose cells and the infiltration of macrophages in adipose tissue were markedly increased after high saturated fatty acids diet or high trans fatty acids diet.4,Expressing of CD68 protein in the viseral adipose was significantly and positively correlated with VW/BW,diameter of the epididymis adipose cell and IRI.To explore the mechanisms of macrophage infiltration and activation in adipose tissue may provide new cellular and molecular targets for prevention and treatment of insulin resistance,type 2 diabetes mellitus and atherosclerosis. |
