The Effect Of Xue Jie Drug-Serum On Apoptosis And 6-keto-PGF_1α,TXB₂ Secretion Of Eutopic Stromal Cell Of Endometriosis

Endometriosis is one of the common, difficult and complicated diseases in gynecology, which is defined as endometriotic tissues growing outside normal uterine cavity. Its incidence has increased considerably in recent years. The main symptom of this disease is gradual dysmenorrhea, and accompanying it, about 30-50% patients can't get pregnant, which has been seriously injuring the patients both in body and mentality. It's pathogenesis remains unclear, then the pathophysiology, treatment programs become a hot research, and Chinese medicine treatment has also become a focus of exploring.There is no disease named endometriosis in Traditional Chinese Medicine. The main symptoms are correlated closely to"dysmenorrheal","abdominal mass","irregular menses"and"sterility"etc. Consider the main pathogenesis is"blood stasis in conception and thoroughfare vessels and uterus", using the method of promoting blood circulation to resolve stasis and resolving abdominal mass.Professor Qian Jing, my director, has accumulated abundant clinical experiences of curing this disease for Twenty-odd years, and she thinks that endometriosis generate the pathological features of silt. Based on these clinical experiences, combining the theories of both traditional Chinese medicine and modern western science of medicine, she invented the promoting blood circulation drug named Xue Jie for curing endometriosis with great concentration. The clinical practice demonstrates that the curative effect of Xue Jie is considerably satisfactory.For probing into the effect and mechanism of curing endometriosis by Xue Jie, we have conducted the laboratory research.Experiment:1,The eutopic stromal cell of endometriosis was cultured by modified method of primary culture. The cell types were identified by immunocytochemical method and the cell morphology was observed by microscope.2,The eutopic stromal cell of endometriosis has been interposed by different dosage drugs of Xue Jie drug-serum and danazol drug-serum, then growth inhibition was detected by MTT and the morphology of apoptosis was observed by dual fluorescent staining.3,The level of 6-keto-PGF1α,TXB2 was determinated in 24h,48h and 72h from supernatant liquid by ELISA.Result:1,The success rate for culture the eutopic stromal cell of endometriosis was 81.40%.Immunohistochemistry showed that the purity of stromal cell was more than 95 %.2,Both Xue Jie drug-serum and danazol drug-serum can inhibit growth and promote apoptosis of the eutopic stromal cell of endometriosis, and danazol has better effect than Xue Jie .3,Xue Jie drug-serum can adjust the function of endocrine with the eutopic stromal cell of endometriosis. Xue Jie drug-serum can lower the level of TXB2, and adjust the balance of TXB2 and 6-keto-PGF1α.Danazol drug-serum had no significant effect.Conclusion:1,This experiment successfully established the eutopic stromal cell model of endometriosis and provides a good experimental model for endometriosis research.2,Xue Jie drug-serum can inhibit growth and promote apoptosis of the eutopic stromal cell of endometriosis.We speculate that Xue Jie induces apoptosis, promotes ectopic foci to shrink and disappear, at the same time it play a preventive role to promote apoptosis of potential lesions of the endometrium and to inhibit the cultivation and growth.3,Xue Jie drug-serum can lower the level of TXB2 and adjusting the balance of TXB2 and 6-keto-PGF1α.The effect of smitigating and treating dysmenorrheal may be achieved by improving the blood rheology, bettering the pelvic blood circulation, inhibiting uterine contraction, relaxing smooth muscle.
