Purpose To study the expression of human mammaglobin (hMAM) and small breast epithelial mucin(SBEM). To explore the correlation between micrometastasis and hMAM,SBEM in the peripheral blood of the patients with breast cancer.Object The SBEM-mRNA,hMAM-mRNA in the peripheral blood of the patients with breast cancer.Methods To uninterrupted dilute the total- ribonucleic acids density of the MCF-7 breast cancer cell , make its density be dilutede by 10 times and reverse transcription the total- ribonucleic acids to complementary deoxynucleic acid. Then amplification the complementary deoxynucleic acid by PCR and Nested PCR to contrast the sensitivity of the two methods . penpheral blood samples were obtained from 92 patients with breast carcinoma ,and 20 patients were with benign breast diseases ,and 10 patients with other system malignant tumor ,and 10 normal healthy volunteers. To identify breast carcinoma cells in peripheral blood, hMAM-mRNA and SBEM-mRNA were amplified from total RNA extracted from wholeblood by nested reverse transcription-polymerase chain reaction (Nested-RT PCR).The relationship between the expression of hMAM -mRNA,SBEM-mRNA and its correlated clinical marker was analysed.Results To identify breast carcinoma cells in peripheral blood Nested-RT- PCR is the most sensitivity, it can amplified 0.005 nanogram RNA extracted from MCF-7 breast cancer.hMAM-mRNA and SBEM-mRNA was not detectable in the blood from normal healthy volunteers and patients with benign breast disease and the patients with other system malignant tumor. Among 92 breast carcinoma patients, 40 were Nested-RT- PCR positive for hMAM -mRNA. The positive rate was 43.5%(40/92). 43 were Nested-RT -PCR positive for SBEM -mRNA. The positive rate was 46.7%(43/92). The expression of hMAM–mRNA and SBEM-mRNA in peripheral blood was not correlate with patient's estrogen or pregnant receptor; menstruation state ; primary tumour size. The expression of hMAM–mRNA and SBEM-mRNA in peripheral blood was correlate with patient' s age; the state of c-erbB-2 and carcino embryonic antigen and carbohydrate antigen15-3.Conclusion Micro metastasis is an independent prognostic factor for breast cancer. hMAM–mRNA and SBEM-mRNA is a special marker of the breast cancer patient' s peripheral blood , hMAM–mRNA or SBEM-mRNA in peripheral blood may be indicator of breast cancer cells ,which might predict hematogenous spreading metastasis of tumor cells in patients with breast cancer. Nested-RT PCR is the sensitivity method to identify breast carcinoma cells in peripheral blood.
