| Objective:To investigate effects of botulinum toxin type A on the expression of TGF-β1and bFGF in hyperplastic tissues of Wistar rats.Methods:50 male Wistar rats were divided into sham operation group(n=10)and model group(n=40).After castrated,40 rats of model group were injected subcutaneously testosterone propionate 50mg/kg.d to induce BPH;10 rats of sham operation group were injected subcutaneously the same dose of NS.After 4 week,the 4 model rats were randomly killed.The structure of prostates were observed by light microscopy.It proved that rat model was successful.The other 36 model rats were randomly divided into high does group,low does group and negative control group.The high,low does group received respectively 10U/0.1ml,5U/0.1ml BTX-A injected into the prostate;negative control group were injected with 0.1ml NS into the prostate at the same position.After 3 weeks all rats were killed and weighed the removed prostate.The expression of TGF-β1 and bFGF in rats of hyperplastic prostatic tissues were analysed by means of immunohistochemical technique.Results:Comparing with negative control group,prostate weight,the positive rate of TGF-β1 and bFGF of high,low dose group were significant difference (p <0.05) : prostate weight decreased,the expression of TGF-β1 increaced and the expression of bFGF decreased.Conclusion:The treatment of BTX-A changed the expression of TGF-β1 and bFGF of prostatic tissues inhibiting cell proliferation and inducing apoptosis. |
