| Objective: To study the effects of active ingredients (emodin and magnolol) in Da Cheng Qi tang(DCQT)on voltage-gated potassium currents of interstitial Cajal cells(ICCs) in small intestine of Wistar rat using the whole-cell patch clamp technique.Methods: The ICCs in small intestine of Wistar rat in SPF level were isolated and cultured. The voltage-dependent K~+ currents in ICCs was recorded by whole-cell patch clamp for observing effects of active ingredients, emodin and magnolol, in DCQT.Results: The maximum amplitude of the voltage-dependent K~+ currents in ICCs using the magnolol were 455.64±10.20 pA which was more 26.38% than the maxium amplitude in control 360.54±28.88 pA(P<0.05); the maximum amplitude using the emodin were 496.30±26.51 pA which was more 37.65% than the maxium amplitude in control 360.54±28.88 pA (P<0.05).Conclusion: The results suggested that the active ingredients emodin and magnolol in DCQT could obviously activate the voltage-dependent K~+ currents in ICCs in rat. DCQT could improve the recovery of gastroinestinal movtility dysfunction in MODS and treat the MODS induced with peritonitis by activating the voltage-dependent K~+ currents in ICCs. |
