| Objective:To evaluate the inhibitory effects and the possible mechanism of endostatin on Human Adenocarcinoma Cells A549 and Human Umbilical Vein Endothelial Cells HUVEC.Methods:1. MTT to observe the inhibitory effects of endostatin with different concentrations(10, 20, 30, 40, 50μg/ml, 48h) and endostatin(30μg/ml) with different time(24, 48, 72h) on the growth of A549 and HUVEC cells.2. Flow cytometry(FCM) to detect the influences of endostatin (30μg/ml,48h) on cell cycle and apoptosis of A549 and HUVEC cells.3. ELISA to assay the influences of endostatin (30μg/ml,48h) on VEGF in Supernater of A549 cells and HUVEC cells.4. RT-PCR to evaluate the influences of endostatin (30μg/ml,48h) on expression of Survivin gene of A549 cells.Results:1. MTT showed that the growth of Human Adenocarcinoma Cells A549 and Human Umbilical Vein Endothelial Cells HUVEC was significantly inhibited (P<0.05) by endostatin(20,30,40,50μg/ml),the inhibitory effects presented time-dose -dependent.2. FCM showed that the cell cycles of A549 and HUVEC cells were inhibited by endostatin (30μg/ml,48h) and the apoptosises of A549 and HUVEC cells were increased(P<0.05) by endostatin (30μg/ml,48h).3. ELISA showed that the secretions of VEGF from A549 and HUVEC ce1ls were inhibited(P<0.05) by endostatin (20,30,40,μg/ml,48h),and it presented dose–dependent.4. RT-PCR showed that the expression of Survivin gene of A549 cells was inhibited(P<0.05) by endostatin (30μg/ml,48h) , compared with normal A549 cells.Conclusion:1. Endostatin can inhibit the proliferation of Human Adenocarcinoma Cells A549 and Human Umbilical Vein Endothelial Cells HUVEC with time-dose -dependent.2. The mechanism of Endostatin,s inhibition on Human Adenocarcinoma Cells A549 and Human Umbilical Vein Endothelial Cells HUVEC may be the cell cycle inhibition and apoptosis.3. Endostatin can inhibit the secretions of VEGF from A549 and HUVEC ce1ls with dose–dependent.4. Endostatin can inhibit the expression of Survivin gene of A549 cells, which is one of the mechanisms related to A549 cells apoptosis. |
