| Objective:To investigate the transcription and expression of S100A4 gene,protein and E-cad in transitional cell carcinoma of the bladder (TCCB) and discuss their significance in carcinogenesis and progression of TCCB,as well as their relations with angiogenesis and prognosis.Methods:1.The expression of S100A4 gene in 38 cases of TCCB and corres- ponding peri-cancerous tissues were examined by reverse transcriptase polymerase chain reaction(RT-PCR).2.The expression of S100A4 protein and E-cad in 38 cases of TCCB and corresponding peri-cancerous tissues were examined by immunohisto- chemical stain.Results:1.The positive rates of S100A4 gene in TCCB and peri-cancerous tissues were 60.5% (23/38) and 0.0%(0/38), respectively. The expression level of S100A4 gene in TCCB was significantly higher than that in normal bladder urothelial tissues(P<0.005).The expression rate of S100A4 gene in TCCB was positively correlated with tumor grade,clinical stag and lymph node metastasis(P<0.05).2.The positive rate of S100A4 protein in TCCB and non-cancerous tissues were 55.3% (21/38) and 0.0%(0/38), respectively.The expression level of S100A4 protein in TCCB was much higher than in peri-cancerous tissues(P<0.05),at the same it was related positively with pathological grade(P<0.05),clinical stage(P<0.05) and lymph node metastasis(P<0.05).3. The positive rate of E-cadherin protein in TCCB and peri-cancerous tissues were 47.4% (18/38) and 81.6%(31/38), respectively.The expression of E-cadherin protein in TCCB,the differences had statistical significance as follow: pathological grade(P<0.05),clinical stage(P<0.05) and lymph node metastasis(P<0.05).The differences had no statistical significance involved:age,sex and tumor size(P>0.05).4.The relationship of the expression of S100A4 protein and E-cadherin protein was analyzed by statistics,results show that it was negative correlation(rs=-0.524,P<0.05).Conclusions:1.The highly positive rate of S100A4 gene and protein in TCCB reveals that S100A4 may play an important role in the carcinogenesis and progression of TCCB. with clinical stage,pathological grade,lymph node metastasis and recurrence in TCCB.It shows S100A4 may possibly become a new marker for diagnosis and estimation of TCCB.3. Expression of E-cadherin protein reduces in TCCB.Decreased expression of E-cadherin are related with malignant progression of TCCB and tumor metastasis.4.Expression of S100A4 protein and loss of E-cad expression are significantly associated with tumor progression in TCCB,they perhaps affect the occurrence,devolopment and metastasis of TCCB together.they are valuable markers predicting biological behavior and trend of metastasis of TCCB.S100A4 may be one of the target in gene therapy of TCCB. |
