| Objective:To study the influence of dys-psychological stress on the mouse ovary function with female mice dys-psychological stress model, and discuss Jin-Shui-Bao's control action to the ovary function.Methods:1. Setting up dys-psychological stress model with restraint, In the operation the stress group BALB/c mice are put in turn in the 50ml centrifuge tube. Restraining the mice 8h/day(10:00PM-6:00AM) for three weeks, 5d/week. 1/2 animals are executed in the first estrus after the stress stimulation stops, striping off the ovary to weigh, and makes the examination. And the remaining 1/2 mice and the male mice (same age homogeneous ) are put into the same cage for12 day (3 estrous cycles).2. The experiment is carried out in 4 groups: (1) control group; (2) stress group; (3) Jin-Shui-Bao group; (4) Jin-Shui-Bao+stress group:3. Observing each group's survival condition, body weight, estrous cycle, ovary (weight, ovary index), ovary tectology change, rate of pregnancy and the filial generation number.4. With ELISA method, examining each group's blood serum cortisone (CORT), tumor necrosis factor alpha (TNF-α), estradiol (E2), progestogen (P), Follicle Stimulating Hormone (FSH) and luteotrop(h)ic hormone (LH) content.Results:1. Successfully establishing the female mice dys-psychological stress model with the fetter method.2. Comparing BALB/C mice weight,ovary weight and ovary index.(1)The stress group doesn't grow well and its eating and activity are not good;(2) The stress group's body weight is lower than that of the control group, and the difference is significant(P<0.01); the Jin-Shui-Bao + stress group's body weight is lower than that of the Jin-Shui-Bao group, and the difference is significant (P<0.05); the body weights of both the Jin-Shui-Bao group and the Jin-Shui-Bao +stress group increase compared to the stress group, and the difference is significant (P<0.01).(3) The stress group's ovary weight and ovary index are lower than those of the control group, the Jin-Shui-Bao group and the Jin-Shui-Bao+stress group, and the difference is significant (P<0.01); the Jin-Shui-Bao + stress group's ovary weight is lower than that of the control group and the Jin-Shui-Bao group, and the difference is significant (P<0.01); the Jin-Shui-Bao + stress group's ovary index is smaller than the Jin-Shui-Bao group, and the difference is significant (P<0.01).3. Comparison of mouse estrous cycle(1) The stress group's estrous cycle is longer than that of the control group and the Jin-Shui-Bao group. (P<0.01);(2) The Jin-Shui-Bao+stress group's estrous cycle is lengthened than that of the control group and the Jin-Shui-Bao group (P<0.05);(3) The Jin-Shui-Bao+ stress group's estrous cycle is shorter than that of the stress group (P<0.05);4. Comparison of mouse blood serum CORT, TNF-α, FSH, LH, E2, and P value.(1) The stress group's CORT and TNF-αare higher than those of the control group and the Jin-Shui-Bao group (P<0.01); stress group's FSH, LH, E2 and the P value are lower than those of the control group and the Jin-Shui-Bao group (P<0.01);(2) The Jin-Shui-Bao + stress group's CORT and TNF-αare higher than those of the control group and the Jin-Shui-Bao group (P<0.05); the Jin-Shui-Bao + stress group's FSH, LH, E2 and the P value are lower than those of the control group and the Jin-Shui-Bao group, (P<0.05).(3) The Jin-Shui-Bao + stress group's CORT and TNF-αare lower than those of the stress group (P<0.05); the Jin-Shui-Bao + stress group's FSH, LH, E2 and the P value are higher than those of the stress group, (P<0.05).5. The mice ovary color turns pale in the stress group , and under the light microscope it is discovered that the total ovarian follicle and the mature ovarian follicle reduce. 6. Comparison of pregnancy rate and filial generation number.(1) The pregnancy rate of the stress group mouse is lower than that of the control group, and the difference is not significant (P>0.05); the stress group's filial generation number is lower than that of the control group, and the difference is significant (P<0.05);(2) The pregnancy rate of the Jin-Shui-Bao + stress group mouse is higher than that of the stress group, and the difference is not significant (P>0.05); the Jin-Shui-Bao + stress group's filial generation number is higher than that of the stress group, and the difference is significant (P<0.05).Conclusion:1. The dys-psychological stress causes the excessive synthesis of mouse cortisone , thus affecting the hypothalamic-pituitary-ovarian axis function, furtherly causing the Follicle Stimulating Hormone (FSH) and the luteotrop(h)ic hormone (LH) secretion to reduce resulting into ovulation barrier, estradiol, progesterone secretion drop and above all, ovary function depression.2. The Jin-Shui-Bao causes the fetter stress mouse weight gain, blood serum FSH, LH, E2, the P level and the filial generation number increases, CORT, TNF-αreductions, which indicates the Jin-Shui-Bao improves the ovary function. |
