Objective:To investigate the effect of the CO2 pneumoperitoneum on the level of serum CRP, TNF-α, IL-6, IL-4, IL-10 in the rats with severe abdominal inflammation in different Post-operative periods, and to explicit the changes in their immune function.Methods:50 male SD rats, divided into 5 group randomly ,10 for each group . GroupI: laparotomy ; Group II: pneumoperitoneum ; Group III: severe abdominal inflammation ; Group IV: pneumoperitoneum control; Group V: laparotomy control. The rat model of severe abdominal inflammation was according to cecal ligation and puncture(CLP) as Wichterman.The Severe abdominal cavity inflammation rats. were laparotomy or treated with CO2 pneumoperitoneum after cecalling ligation and punctureing (CLP) 16 hours.The blood samples were gathered for examing C-reactive protein(CRP), tumor necrosis factor-α( TNF-α), IL-6, IL-4, IL-10 before they were treated and after they were treated about 8 hours, 24 hours, 48 hours, 72 hours.Results:The experimental results were as follow: In Group I,Grou II,Group III,serum CRP,TNF-α,IL-6,IL-4 and IL-10 were significantly higher than in Group IV and Group V before and after the were treated(P<0.01). Serum CRP, TNF-α, IL-6, IL-4 and IL-10 in Group II were significantly lower than that in GroupI in postoperative(P <0.01). Serum CRP,TNF-α,IL-6,IL-4 and IL-10 in GroupI were significantly higher than that in Group III in postoperative (P <0.01); they were higher in Group II than in Group III in postoperative, but there was no significant statistical difference (P >0.05).Conclusion:(1)Surgery could lead to the secondary injury , and make theSIRS and CARS becoming more serious to severe abdominal cavity inflammation rats. (2) After operation in severe abdominal cavity inflammation rats ,the extent of in?ammatory response and anti-in?ammatory response in CO2 pneumoperitoneum group were significantly lower than in laparotomy group. (3) CO2 pneumoperitoneum could better preserve the balance of in?ammatory response and anti-in?ammatory response on Severe abdominal cavity inflammation rats.CO2 pneumoperitoneum can better protect the immune function than laparotomy on Severe abdominal cavity inflammation rats...
