ObjectivesTo study the E-cadherin monoclonal antibody SHE78-7 enhance HCE1(cervical cancer cell line ) Multicellular Spheroids's sensitivity to Cisplatin and its relationship with E-cadherin expression.Methods(—) To establish HCE1 multicellular spheroids model using liguid overlay technique and rotating culture technique。(二) Observation E-cadherin monoclonal antibody SHE78-7' role in HCE1 multicellular spheroid morphology change。(三) before and after SHE78-7 blocking cell adhesion,Detection different concentrations of cisplatin on HCE1 multicellular spheroid inhibition of proliferation with GST-1 method,and single-cell as a control, calculate IC50 values,multicellular resistance index MCRI,and mapping the concentration-inhibition curve.(四) before and after SHE78-7 intervention HCE1 multicellular spheroid,detection E-cad expression with Immunocytochemical method,and monolayer cells as controls.Results(一) establishment of HCE1 multicellular spheroid model:we observed the growth of HCE 1 monolayer cells were in a good condation, after 24h in rotating culture,cells began to aggregated into multicellular spheroids,and becoming more and more compact and larger in size relay to culture time.2.histological structure of HCE1/MCS:central necrosis core and cell outgrowth around the MCS were observed when MCS adherented.Non-structure red-stain region was observed using HE stainning.3.Growth curve was drawed:be cultured till 10th days,the growth of HCE1/MCS turned into platform,the double time of HCE1/MCS cells is 4.43±0.47 days.(二) E-cadherin monoclonal antibody SHE78-7 on cell-cell adhesion inhibition:Monolayer with SHE78-7:loose connections between cells and can not grow into a film,control group:with close connections between the cells,into a film growth;multicellular spheroid with SHE78-7 group: can not form multicellular spheroid,or cells together,but loose structure form while control group with Stable structure。(三) before and after SHE78-7 blocking cell adhesion,Detection inhibition of proliferation with different concentrations of cisplatin on HCE1 multicellular spheroid with GST-1 method,while single-cell as a control:before SHE78-7 blocking cell adhesion:the cisplatin' IC502D in the single-cell:6.04mg/ml,while IC503D in the multicellular spheroid: 18.32mg/ml,MCRI:3.03.after SHE78-7 blocking cell adhesion:the cisplatin' IC502D ' in the single-cell:3.17 mg/ml,while IC503D'in the multicellular spheroid: 6.96mg/ml,MCRI ':2.19. We can see that,multicellular resistance index decreased significantly after SHE78-7 blocking cell adhesion。(四) before and after SHE78-7 intervention HCE1 multicellular spheroid,detection E-cad expression with Immunocytochemical method,and monolayer cells as controls:positive staining showed brown particles,the main expression of single-cell membrane,multicellular spheroid membrane,cytoplasmic。①the expression of E-cad in HCE1 Multicellular Spheroids increased significantly than monolayer(P<0.05)②SHE78-7 can't change E-cad expression in HCE1 single or multicellular spheroids(P>0.05)。Conclusion1.anti-E-cadherin monoclonal antibody SHE78-7 can inhibit the formation of HCE1 multicellular spheroid.2.SHE78-7 can enhance the HCE1 multi-cellular spheroid' Sensitivity to cisplatin.3.SHE78-7 can effectively inhibit the formation of HCE1 Multicellular Spheroids,but E-cad expression in HCE1 Multicellular Spheroids have little change。...
