| AIM To investigate the effects of NF-κB in experimental model of endotoxemia induced by LPS and to provide theoretical basis use as a therapeutic target.Methods Adult SD rat endotoxemia model was established via tail vein administration of LPS and verified by altered liver function and histology.The animals were randomly divided into groups to receive treatment with different doses of PDTC,then studied the influences on spleen NF-κB P65 expression level studied histochemically.Results Positive spleen cell nucleus NF-κB P65 activation expression began to appear at lh time point after LPS challenge and reached peak at 3h time point and then gradually declined thereafter,at the 24-h time point the difference between control group became insignificant.The effect was dose dependent,the 200mg/kg group showed the most prominent inhibition of NF-κB P65 expression as well as inhibition of TNF-αexpression.Conclusion The LPS induced rat experimental endotoxemia liver and spleen experienced activation of NF-κB P65 as well as TNFα,the antioxidant PDTC significantly suppressed the activation expression of both NF-κB P65 and TNFαin a dose-dependent manner. |
