| Objective: To investigate the expression of the AI(apoptosis index )and FasL(factor associated suicide ligand) of cultured human cytotrophoblast cells under EmAb(antiendometrium antibody) environment.Methods: 5ml EmAb positive ,other immuno-relative antibody negative blood of RSA(recurrent spontaneous abortion) patient were collected. 5ml normal women blood ,steted, centrifugated were collected and stored at-80°C for use. Chorionic villi tissue of normal 6-8 week pregnancy were obtained. Cytotrophoblast cells were collected by Trypsin-DnaseI digestion and Percoll gradient centrifugation . CTs were primarily cultured and their purities were accessed by immunocytochemistry. Secondary cultured CTs were cultured under EmAb(+) serum group and EmAb(-) serum group for 0h,48h,72h,96h. Typan blue exclusion test was used to observe the vigor and calculate the viability of CTs. Observe AI by TUNEL andFasL expression by immunohistochemistry at every culture schedule.Results: AI in EmAb (+) serum group was significantly higherthan that of EmAb(-) serum group (P<0.05) .FasL concentrations in EmAb(+) serum group were significantly lower than those of EmAb (-) serum group when measured at 72h, 96h culure period. AI and FasL concentrations were not obviously different when measured at 48h culure period (P>0.05) . Conclusions: AI of cytotrophoblast cells increased and FasL expression decreased when they were cultured under free EmAb environment.Significance:AI and FasL expression of CTs at different culture environment help to understand the etiology of RSA. AI of the EmAb(+) serum group was significantly higher than that of EmAb(-) serum group (P<0.05) . This helps to investigate EmAb behavior in apoptosis mechanism of RSA. FasL expression of CTs of EmAb(+) serum group at 72h, 96h culure period was significantly lower than that of EmAb(-) serum group. There were no lymphocytes in pured CTs. They can't make Fas(+) cells die . It concludes that apoptosis of CTs in this experiment has nothing to do with FasL expression but has something to do with it in internal environment. Decreased FasL Expression of EmAb(+) RSA patient can decrease Fas(+) suiccide lymphocyte apoptosia of mother blood and deciduas. Fas(+) suiccide lymphocyte increase cells in placenta apoptosis , so spontaneous abortion occurs. |
