To study the effect and possible mechanism of alcoholic exactraction of Semen Cuscutae(AESC) on cell cycle and DNA damage of aging model rats. Method: 50 Wistar rats were divided into young (10), aging(10), three different administration groups of 15days, 30 days and 45 days(30) randomly.Aging model rats were made by injected D-gal (48mg/d) on the nape 45 days. After that, the administration groups begin to be drenched by stomach with alcoholic extraction of Semen Cuscutae at a dosage of 0.8g/kg/d. Kill the rats after administration of 15 days,30 days and 45 days respectively. RT-PCR method was used to examine the mRNA expression of p16,p21,cyclinD1,cyclinE and PCNA; Immunohistochemistric method was used to measure the protein expression of P16,P21,CyclinD1,CyclinE and PCNA. To assay the DNA damage in aging model rat spleen lymphocytes by the single cell gel electrophoresis (SCGE) technique; Observe the effect of administration of different days on these indexes in comparison.Result: Compared with young group, the aging group has several changes, the expression level of mRNA and protein of p16,p21 and cyclinD1were increased(p<0.01); The expression level of mRNA and protein of cyclinE and PCNA is decreased (p<0.01); the percentage of cell with comets of spleen lymphocytes increased (p<0.01). The AESC could decrease the expression level of mRNA and protein of p16,p21,cyclinD1(p<0.01), decrease the percentage of cell with comets of spleen lymphocytes (p<0.01), could increase the expression level of Mrna and protein of cyclinE and PCNA.Conclusion: The AESC can change the expression of cell cycle factor, decrease the DNA damage, so it plays role in delaying aging. It effect depends on the length of administration.
