To investigate the effect of semen,seminal Plasma and PMSG+hCG on endometrial receptivity in the rabbit by determing the expression of endometrial pinopodes and the level of endometrial interleukin-6(IL-6).The aim of this study is to find a new method improving implantation rate.Totally 60 healthy adult California female rabbits were randomly assigned to four groups:superovulation,semen,seminal Plasma and control group with 15 rats in each group. The superovulation group were given subcutaneous injection with 80IU/PMSG at proestrus and ear vein injection with 100/hCG at three days later,then didn't mated with male rabbits; the semen group received bilateral tubal ligation,then one month later after inducing equivalent amount of Physiological saline in the same way mated with intact males at proestrus;the seminal plasma group after inducing equivalent amount of Physiological saline in the same way mated with males one month after vasectomized;and the control group after inducing equivalent amount of Physiological saline in the same way didn't mated with male rabbits.The three groups were sacrificed at 5th,6th,7th,8th,9th day and taken uterine specimens respectively.The expression of endometrial pinopodes was determined dy scanning electron microscopy and endometrial levels of IL-6 were measured by enzyme-linked immunosorbant assay(ELISA).Result:1.The expression of Pinopodes in endometrial surface in each group:semen and seminal Plasma groups were superior to control group,control group was superior to superovulation group,but there is no significantly difference between semen and seminal plasma group.2.the changes of the level IL-6 in endometrium in all groups were similar,it increased obviously from day 6 and reached the peak on day 7 which was about twice of that of day 5 and then decreased gradually,but the levels of day 8 and 9 were still higher than that of day 5.the level of IL-6 in groups of seminal plasma and semen were significantly higher than in control group and superovulation group(P<0.01);control group were significantly higher than in superovulation group(P<0.01);and there is no significantly difference between Semen and seminal plasma group(P>0.05).Conclusions:1.seminal plasma could obviously improve pinopodes and IL-6 expression of endometrium in the implantation period in rabbits.2.PMSG+hCG could significantly reduced the pinopodes expression in endometrial surface,make it growth carry forward and extinction advanced.PMSG+hCG could also decreased IL-6 expression of endometrium in the implantation period in rabbits.3.Seminal plasma can enhance the endometrial receptivity,and also can augment the embryo plantation rate,which is expected to be applied in clinic with improving the pregnancy rate of embryo transfer.4.It should be paid attention to that the superovulation adversely affects the endometrial receptivity,and the impact of the mechanism of it is not entirely clear,which still need to be further researched and discussed.
