| Objective The removal of calculus , decrease and prevention of the accumulation of plaque is the key to the treatment and prevention of recurrent of periodontal disease. The most known effective way is mechanical removal of plaque and calculus. The maintenance needs the patient's own plaque control. But some type of periodontal disease still need the use of drug treatment as adjunct therapy. Recently there are a lot of researches being done on the treatment of periodontal disease using medicine. And have found several periodontal drug. But there are still disadvantages. this research hope to design a compound medicine that can effectively treat local periodontal disease without the forming of drug-resistance effect.Methods The theory is based on the oxidative effect of Methylene Blue.which let the other main component Minocycline that originally absorb light easily can extend its bactericidal effect. Since Methylene Blue is the receptor during the process of oxidation and reduction process. By the combination of the blue color of Methylene Blue and yellow color of Minocycline that formed green color , and with the absorption peak of Methylene Blue (609-668nm), which have the effect of both slowing down of the oxidative effect of Minocycline and the prevention of the destruction by light .Also Methylene Blue has also an effective bactericidal effect on Gram positive bacterial, and Minocycline has bacteriostatic effect on Gram negative effect. Then use of this two component has additional effect , and can more effectively suppress the growth of bacterial in periodontal pocket, and can achieve the objective of periodontal treatment.Results In animal experiment, the control group(group A),group B with the use of the slow releasing pocket delivery Minocycline compound as experimental group. Each have 12 experimental rat and have the total bacterial count. The result show in the control group (group A) has no significant change form day 0 to day 7. Experiment group(group B) showed the decrease of total bacterial count, and has significant statistical meaning (p<0.005:0.001),the microorganism population also has changed in the periodontal pocket, Aggregatibacter actinomycetemcomitans, Porphyromonas gingival is, spirochete, Gram positive mobile bacteria has significantly reduced or disappeared, Gram positive coccid and mobile rods increase .microscopically, the control group showed significant inflammatory cell over the localized ginigival tissue. The experimental group(group B),after 48 hours of application of the compound medicine showed significant improvement, and showed no inflammatory effect after 7 days. This showed that the application of the compound medicine has bactericidal effect over the periodontal disease and can eliminate completely the inflammatory effect.Conclusions The compound medicine has bactericidal effect over the periodontal diseases. |
