Experimental Study On Pulmonary Fibrosis Treated With Total Glucosides Of Danggui Buxue Decoction In Rats

Pulmonary fibrosis(PF)is a kind of serious lung diseases and is the common final of many pulmonary diseases. It is as diffuse exudation of pulmonary interstitial ,infiltration and fibrosis as the main lesion. the process involves inflammatory damage of lung tissue, the movement of inflammatory cells, release of inflammatory mediators, fibrosis and cicatrices caused by the mediation of injury and disorder of tissue restoration.However the mechanism is still unclear to us and there is no satisfactory drug therapy for it. Glucocorticoids is the first choice in treating PF nowadays,yet many studies indicated that it could induce some serious diseases such as osteoporosis and so on.Traditional Chinese medicine has a bright future in preventing and treating PF and many have made great progress in clinical therapy.Danggui Buxue Decoction is a classical Chinese medicine Formula, composed of Astragalus and Angelica,has efficacy of Replenish qi and hematosis, support healthy energy.some research indicates that the formula has anti-oxidation effects. There are many components in Danggui Buxue Decoction and total glucosides of Danggui buxue Decoction is a kind of yellow powder extracted from Danggui buxue Decoction in our institute ,which is tested and main active ingredient is astragaloside and ferulic acid. Astragaloside and ferulic acid have been proved to be good effect to pulmonary fibrosis. so we adopted in this experiment to observe the effect of total glucosides of Danggui Buxue Decoction on bleomycin A5 induced pulmonary fibrosis in rats and to explore its pathogenesis. we copy the fibrosis model by infusing bleomycin(5mg/kg)through tracheal intubation,choose prednisone as the positive drug. On that base we study the preventive effects of total glucosides of Danggui buxue Decoction(TGDB) on pulmonary fibrosis induced by bleomycin A5 and it's mechanismsof action.The main contents are divided into two sections follows.1. preventive effects of TGDB on pulmonary fibrosis induced by bleomycin A5compared with model control group, TGDB can significantly reduce rats'coefficient lung, content of lung tissue Hyp and Serum MDA, HA, LN, CⅢlevels, increased GSH-Px, SOD levels.Histopathological examination and morphological analysis of semi-quantitative results also show that TGDB can reduc the degree of pulmonary fibrosis significantly, and reduce inflammatory reaction. results suggest that TGDB has positively preventive and protective effects on bleomycin A5 induced rat pulmonary fibrosis.2. the mechanisms of TGDB prevent composite factors indused pulmonary fibrosis in ratsTGDB can significantly reduce serum TGF-β1 level,and RT-PCR results also showed that TGDB significantly reduced the TGF-β1, Smad4 expression in rats'tissue compared with the model control group, which suggested the preventive effects of TGDB on bleomycin A5 induced pulmonary fibrosis might be suppress the elevated expression of TGF-β1 and Smad4.
