| Objective:Approach the regulation and sense of change of CD4+CD25+Treg cell and CD4+T cell and CD8+Tcell in the invasion and pathogenetic condition of leukemia, through researching cell ratio and change regularity of CD4+CD25+Treg cell and CD4+T cell and CD8+Tcell in the peripheral blood before and after chemotherapy. Research the relation between the invasion and development of leukemia and them, and provide foundation to the invasion and immunity treatment of leukemia.Methods:Detected cells by FCM of micro amounts whole human blood. Detect labeling surface antigen of lymphocyte with staining of immune of luorescence labeled with three colors. Analyze data with cellquest software. Classify T cell and normal cell mass with setting up gate labeled with fluorescence CD3, and record the percent of positive cells and diminish the value of non-specificity control. Evaluate the expression level of CD4+CD25+Treg cell with the ratio of CD4+CD25+Treg cell to CD4+T cell in the peripheral blood, in order to further eliminate the examination effect posed by leukemia cell in the samples of leukemia patients.Statistical assay:The date was dealt was dealt by SPSS 17.0 statistical software. Experimental date was indicated by the median (p25-p75).The relevance of using non-parametric Spearman correlation coefficient analysis. a=0.05 is the size of test.Results:1 Normal subjects and patients with peripheral blood CD4 + T cells, CD8 + T cells, CD4 + CD25 + regulatory T cells compared1.1 Peripheral blood CD4 + T cells compared: Normal human peripheral blood CD4 + T cells was 49.0 (40-56.25), in patients with pre-treatment CD4 + T cells was 48.0 (33-59), Z= -0.30, P= 0.98, P>0.05,resulting in no statistical significance.1.2 Peripheral blood CD8 + T cells compared: Normal human peripheral blood CD8 + T cells was 38.5 (35-52.75), pre-treatment of patients with CD8 + T cells was 38.5 (28-43.5), Z =-0.78, P =0.43,P>0.05, resulting in no statistical significances.1.3 Peripheral blood CD4 + CD25 + Treg cells compared: Normal human peripheral blood CD4 + CD25 + Treg cells was 3.53 (2.11-4.36), per-treatment of patients with CD4 + CD25 + Treg cells was 7.71 (4.61-10.87), Z =-5.54, P< 0.00, resulting in statistical significance.2 Before and after treatment in patients with peripheral blood CD4 + T cells, CD8 + T cells, CD4 + CD25 + Treg cells compared.2.1 Peripheral blood CD4 + T cells compared: Before treatment was 48.0 (33-59), after treatment for 27.5 (22.25-38.25), Z= -4.16, P< 0.00, the results were statistically significant.2.2 Peripheral blood CD8 + T cells compared: Pre-treatment 38.5 (28-43.5), after treatment for 25.0 (16.25-35.25) Z= -3.24, P <0.00, the results of statistical significance2.3 Peripheral blood CD4 + CD25 + Treg cells compared: Pre-treatment to 7.71 (4.61-10.87), after treatment 1.78(1.0-2.24), Z= -4.78, P < 0.00, the results were statistically significant.3 Peripheral blood CD4 + T after treatment than before treatment dropped 42.7%, CD4 + CD25 + Treg cells decreased 76.91%, CD4 + CD25 + Treg cells decreased more significantly.4 Peripheral blood CD8 + T cells and CD4 + CD25 + Treg cells was used to analyze the relevance of non-parametric correlation coefficient Spearman, r=0.379, P =0.076, P>0.05, resulting in no significant correlation coefficient.Conclusion:1. Leukemia in patients with primary peripheral blood CD4 + CD25 + Treg cells was significantly higher than normal control. It Prompted that CD4 + CD25 + Treg cells clearly related with leukemia incidence and the evolution of the disease。It also proved that the existence of T cell in leukemia patients with the quantity and quality of abnormal and subgroup imbalance.2. With improvement of leukemia patients after treatment, the proportion of peripheral blood CD4 + CD25 + Treg cells has declined. It prompt that monitoring of peripheral blood CD4 + CD25 + Treg cells and the ratio of expression level changes, may be the diagnosis and treatment of leukemia to determine the effect of guiding significance to a certain extent.3.The peripheral blood of Leukemia in patients CD8 + T cells and CD4 + CD25 + Treg cells, no significant correlation shows that clinically relevant CD4 + CD25 + Treg cells to CD8 + T cells in vitro for further study. |
