| PART 1 PREARATION OF THE NANOSCALE PERFLUOROCARBON MICROPHERE AND RABBIT VX2 LIVER TUMORObjective: The purpose of this study was to prepared a kind of nanoscale targeting loading-hematoporphyrin ultrasound contrast agent,detected their physical properties and studied the concentrate visualization in viro.Methods: (1) The NPMS,NSPM, NSHM was prepared by high-speed heterogeneity.The microsphere were studied for size, zeta potential , The NSHM were studied for stabilization , encapsulation efficiency and ultrasound imaging in vitro. (2) The rabbit VX2 liver tumor models were made by transplantation method, these were observed by ultrasound, ultrasound contrast agent and pathological sections and evaluated the contrast sonograms by DFY-Ⅱ(ultrasound imaging quantitative analysis software)Results: (1) The NSHM with stable properties had droplet size of (499.1±69.8.)nm, zeta potential of +(14.9±2.9)mV,encapsulation efficiency of (90.6±1.6)% . The lipospheres precipitation could enhanced ultrasound image in vitro To compare with NPMS,NSPM was no significant difference in size (2) The rabbit VX2 liver tumor models were confirmed to make success by ultrasound, ultrasound contrast and pathology method, the microbubbles could significantly enhance the tumor blood vessels and tumor imaging.Conlusion: (1) The mad-self NSHM are consistent to the standard of ideal new targeting loading-drug ultrasound contrast agent,which could be as small droplet size, stabile physical property ,higher encapsulation efficiency and enhanced ultrasound image in vitro(2)The microbubbles can enhanced the sensitivity of the tumor blood vessels and tumors.PART 2 THE APPLICATION STUDY OF THE NSHM IN RABBIT VX2 LIVER TUMORObjective: The purpose of this study was to evaluated targeting and therapeutic action of the self-made NSHM in the rabbit VX2 liver tumor.Methods: (1) 9 rabbit VX2 liver tumor models were randomly divided into 3 groups as follows: NPMS, NSPM and NSHM group Each rabbit were injected into 1ml microsphere,then the rabbit VX2 liver tumor were detect by ultrasound , pathological sections , fluorescence confocal microscopy ,electron microscopy and evaluated the contrast sonograms by DFY-Ⅱ; (2) 6 rabbit VX2 liver tumor models were randomly divided into 2 groups as follows: NPMS and NSHM group. Each rabbit of the NSHM group was injected 1mg/Kg hematoporphyrin. Each rabbit of the NPMS group were injected into the same amount of normal saline (NS), Each rabbit was gived twice 1.5w/cm2 ultrasound irradiation for 3 minutes,then observe the changes in the tumor and calculate the inhibitory rate. Results: (1) The NSPM group and NSHM group could enhance tumor imaging and located in the tumor site,this also was confirmed that the microsphere with sulfatide could locate in the tumor site by pathology, electron microscope and fluorescence confocal microscope.(2) The NSHM group had necrosised in tumor side after ultrasound irradiation after 24 hours.Their inhibitory rate was more than control group.Conlusion: (1) The mad-self NSPM,NSHM could target to tumors image.(2) The NSHM could inhibit tumors growth after ultrasound irradiation... |
