Objective To observe the permeability of albumin and the expression of gap junction protein (Connexin43) of cultured human renal tubular epithelial cells(HKC) which was stimulated with high glucose and insulin,analysis the relationship of the permeability of HKC and expression of Connexin43 .Method The normal HKC were randomized into 6 groups 1.normal control group; 2.high glucose group (30 mmol / L); 3. group of low concentration insulin (10-9 mol / L); 4 .group of high concentration insulin(10-7 mol / L) ; 5.group of glucose (30mmol / L) and low insulin(10-9 mol / L); 6.group of glucose (30mmol / L) and high insulin(10-7 mol / L),the the permeability of albumin in different groups was examinated with Transwell cab,to detective the expression of Connexin43 of HKC in different groups. The expression of Connexin43 mRNA and protein was detected by RT-PCR and Immunity fluorescence .Observe whether there is relevance between above datas.Result Insulin or high glucose stimulated HKC ,up-regulated the cell permeability (P<0.01),and down-regulated the Connexin43 protein expression .The high glucose stimulated the HKC, then add different concentrations of insulin,the permeability of single-layer tubular epithelial cells decreased,and the expression of Connexin43 increased(P<0.01). Analysis whith linear correlation,there was a negative correlation between the expression of Connexin43 protein of HKC and the cell permeabilityConclusion 1.High glucose or insulin stimulated renal tubular epithelial cell , cell permeability increased and Connexin43 expression reduction.2.The effection of high glucose on renal tubular epithelial cell permeability and expression of Connexin43 can be antagonisticed by higher concentrations of insulin.3.The permeability of HKC and the Connexin43 expression is relevant.The changes of Gap junction protein expression in HKC may be an important mechanism for changes in the permeability.
